Impact of Testosterone on Alzheimer's Disease

Vittorio Emanuele Bianchi

Summary: Alzheimer's disease is a neurodegenerative disease influenced by sex hormones. Animal models have shown that testosterone can protect neurons by reducing amyloid-beta production, improving synaptic signaling, and preventing neuronal death. Studies have found that prostate cancer patients receiving androgen deprivation therapy have a higher incidence of dementia and Alzheimer's disease. Testosterone therapy in hypogonadal men with Alzheimer's disease and cognitive impairment has shown some positive results.

TDMQ20, a Specific Copper Chelator, Reduces Memory Impairments in Alzheimer's Disease Mouse Models

Jie Zhao et al.

Summary: Regulation of redox metal ions, such as copper, presents a promising therapeutic approach for Alzheimer's disease. Specific copper(II) chelators like TDMQ20 show potential in improving cognitive and behavioral impairments in various AD mouse models, as well as reducing oxidative stress and enhancing memory function. The positive pharmacological results obtained from different AD models suggest the efficacy of TDMQ20 in treating AD-related symptoms and memory deficits.

Dietary Habits, Selenium, Copper, Zinc and Total Antioxidant Status in Serum in Relation to Cognitive Functions of Patients with Alzheimer's Disease

Katarzyna Socha et al.

Summary: The study examined selenium, copper, zinc, and total antioxidant status (TAS) in the serum of patients with Alzheimer's disease (AD) compared to healthy individuals. Lower concentrations of selenium, zinc, and TAS, as well as a higher copper:zinc ratio, were observed in the serum of AD patients. Dietary habits were found to have a significant impact on the concentrations of these parameters in the serum of individuals with AD.

The role of glutamate oxaloacetate transaminases in sulfite biosynthesis and H2S metabolism

Anna-Theresa Mellis et al.

Summary: Molybdenum cofactor deficiency and isolated sulfite oxidase deficiency are rare genetic disorders caused by impairment of the mitochondrial enzyme sulfite oxidase. Investigation of the contributions of cytosolic and mitochondrial glutamate oxaloacetate transaminase (GOT) in cellular extracts revealed that the cytosolic isoform GOT1 is primarily responsible for sulfite production. Loss of sulfite oxidase activity leads to accumulation of sulfite, H2S, and persulfidated cysteine and glutathione, suggesting an increase in SQR protein levels. Surprisingly, under conditions of sulfite toxicity, none of the known H2S-producing pathways were found to be upregulated, indicating an alternative route of sulfite-induced shift from oxidative to H2S dependent cysteine catabolism.

Neuroprotective Roles of the Reverse Transsulfuration Pathway in Alzheimer's Disease

Bindu Diana Paul

Summary: The reverse transsulfuration pathway is a central hub integrating the metabolism of sulfur-containing amino acids and redox homeostasis, disruption of which may have deleterious consequences and is linked to Alzheimer's disease. Potential nodes of therapeutic intervention discussed in this review may pave the way for the development of novel treatments.

Summary: This study found that the polygenic risk score for Alzheimer's disease and the presence of the APOE ε4 gene were associated with Aβ misfolding, while the Aβ polygenic risk score was not. All genetic markers could predict Alzheimer's disease diagnosis, but Aβ misfolding had higher predictive accuracy than the other genetic markers.

Summary: This paper discusses the potential etiological role of copper imbalance in Alzheimer's disease, suggesting that it may exacerbate neurodegeneration and protein misfolding as a pathogenic mechanism. Integration with genetic risk factors, metabolic abnormalities, neuroinflammation, and other factors is crucial for a comprehensive understanding of the disease.

Copper Imbalance in Alzheimer's Disease: Meta-Analysis of Serum, Plasma, and Brain Specimens, and Replication Study Evaluating ATP7B Gene Variants

Rosanna Squitti et al.

Summary: Evidence suggests that patients with Alzheimer's dementia exhibit disturbances in copper homeostasis. A meta-analysis of studies on copper biomarkers in brain specimens and serum/plasma samples revealed decreased copper in AD brains, increased copper and nonbound ceruloplasmin in serum/plasma, with excess copper associated with a higher risk of AD. Replication study confirmed these findings and indicated a subset of AD patients carrying a genetic haplotype associated with copper imbalance and elevated nonbound ceruloplasmin.
