Zingerone Modulates Neuronal Voltage-Gated Na+ and L-Type Ca2+ Currents

Ming-Chi Lai et al.

Summary: In this study, the effects of Zingerone (ZO) on ionic currents in pituitary tumor cells and hippocampal neurons were investigated using patch clamp technology. The results showed that ZO had varying effects on the amplitude, gating, and hysteresis of ionic currents, which may significantly impact the functional activities of neuronal cells.

Modeling Epilepsy Using Human Induced Pluripotent Stem Cells-Derived Neuronal Cultures Carrying Mutations in Ion Channels and the Mechanistic Target of Rapamycin Pathway

Octavia Yifang Weng et al.

Summary: Epilepsy is a neurological disorder caused by an imbalance of brain excitation and inhibition. Mutations in ion channels and mTOR signaling molecules are associated with this imbalance. The early stage of brain development is crucial in understanding the mechanism of epilepsy. Using hPSCs-derived neural cultures to study epilepsy is an important avenue of research.

The autoantibody-mediated encephalitides: from clinical observations to molecular pathogenesis

Sudarshini Ramanathan et al.

Summary: The autoimmune encephalitis (AE) syndromes are characterized by the presence of autoantibodies targeting specific neuroglial antigens, with clinical phenotypes closely associated with the respective autoantibodies. Early and accurate detection is crucial for prompt initiation of immunotherapy and improved outcomes. This review outlines molecular observations, diagnostic methodologies, clinical phenotypes, therapeutic paradigms, and potential mechanisms of autoimmunity in AE.

Clinical features which predict neuronal surface autoantibodies in new-onset focal epilepsy: implications for immunotherapies

Ronan N. McGinty et al.

Summary: This study aimed to identify surface-directed autoantibodies in adults with new-onset focal epilepsy and evaluate the value of immunotherapy in this setting. The results showed that patients with detected autoantibodies but without encephalitis had excellent outcomes without immunotherapy, suggesting that immunotherapy administration should be guided by clinical features of encephalitis rather than autoantibody positivity. The findings indicate that seizure syndromes with autoantibodies largely fall under the category of autoimmune encephalitis in this cohort.

Clinical spectrum of high-titre GAD65 antibodies

Adrian Budhram et al.

Summary: This study investigated the clinical manifestations, immunotherapy responsiveness, and outcomes of GAD65 neurological autoimmunity. Results showed that patients with high-titre GAD65 antibodies presented with a range of core and secondary manifestations, with stiff-person spectrum disorders being the most responsive to immunotherapy and epilepsy being the least responsive. Complete immunotherapy response was rare, and predictors of poor outcomes included a serum GAD65 antibody titre >500 nmol/L and the presence of cerebellar ataxia.

Association of Rituximab Use With Adverse Events in Children, Adolescents, and Young Adults

Casey Lee McAtee et al.

Summary: Rituximab, a commonly used immunotherapy in pediatrics, is well tolerated among young people with few serious adverse events. However, infections are common, leading to a prolonged period of B cell count recovery often lasting for longer than a year. Further strategies to prevent infections following rituximab should be explored.

The Roles of Calmodulin and CaMKII in Cx36 Plasticity

Georg R. Zoidl et al.

Summary: The translation highlights the initial belief that gap junctions were limited to invertebrates and nonmammals, but recent studies have shown that electrical communication is common in the mammalian central nervous system. The subsequent progress revealed the vital role of gap junction protein Cx36 and its paralogs in forming electrical synapses in mammals and fish, governing the collective activity of coupled neurons with adaptive plasticity. These electrical synapses also orchestrate synchronized neuronal network activity and rhythmic oscillations underlying processes like memory and learning.

Calmodulin Interactions with Voltage-Gated Sodium Channels

Xin Wu et al.

Summary: Calmodulin (CaM) is a small protein that serves as a ubiquitous signal transducer, regulating neuronal plasticity, muscle contraction, and immune response. It interacts with ion channels and plays regulatory roles in cellular electrophysiology. Mutations in CaM-binding IQ domain can lead to various diseases.

Epilepsy in the mTORopathies: opportunities for precision medicine

Summary: The mechanistic target of rapamycin signalling pathway plays a crucial role in regulating cell activities, with pathogenic gene variants potentially leading to epilepsy and neurodevelopmental disorders. Diseases like tuberous sclerosis complex and focal cortical dysplasia type II are associated with excessive activation of the rapamycin pathway.
