Connexons Coupling to Gap Junction Channel: Potential Role for Extracellular Protein Stabilization Centers

Laszlo Heja et al.

Summary: This study discovered subtype-specific protein stabilization centers (SCs) in Cx GJC, and identified the structure of the GJ interface. The unique trans-GJ SC pattern was found to play a major role in stabilizing the GJC complex. Additionally, clusters of SC patterns close to the GJ interface domain were revealed to orient the interface formation.

Over-activated hemichannels: A possible therapeutic target for human diseases

Mauricio A. Retamal et al.

Summary: Intercellular communication mediated by connexins, transmembrane proteins that form two types of channels, plays a crucial role in coordinating cellular responses. While hemichannels normally release signaling molecules in a controlled manner, over-activated hemichannels can exacerbate symptoms in certain pathological conditions. Recent research efforts have focused on developing tools such as small molecules, antibodies, and mimetic peptides to modulate over-activated hemichannels for potential treatment of human diseases.
