Missense mutations can disrupt protein folding and lead to disease, but some mutations are kinetically delayed to prevent interference with wild-type function. The involvement of chaperones, kinetic folding delays, and oligomerization dynamics were further explored in tumor-derived mutants of the tumor suppressor p53, with findings suggesting temperature sensitivity and Hsp90 regulation.
