Packing Preferences of Chalcones: A Model Conjugated Pharmaceutical Scaffold

Summary: By analyzing 232 crystal structures of chalcones, it was found that although pi conjugation would favor a planar molecule, there are a significant number of nonplanar structures. The relative orientation of dimer molecules in the crystal structures shows similarities with the structures generated through CSP. There is no specific dimer geometry associated with preferred lattice energies for the chalcone core, and face-to-face pi stacking is less common. The most common one-dimensional motif found in experimental crystal structures is a side-to-side packing.

One important application of crystal engineering is in the design of pharmaceutical cocrystals, which can systematically tune the physicochemical properties of drugs without altering their molecular structure. This review traces the development of supramolecular synthons as robust hydrogen bond patterns for pharmaceutical cocrystal design, and discusses recent advancements in the field that have led to the launch of several drugs with improved efficacy and bioavailability. The importance of pharmaceutical cocrystals lies in their ability to bridge the gap between drug discovery and development, and their potential to provide high-value pharmaceuticals with economic and environmental benefits.
