Cytotoxic mechanisms of doxorubicin at clinically relevant concentrations in breast cancer cells

Rachel E. Nicoletto et al.

Summary: This review updates the concentration and time-dependent trends of doxorubicin (DOX) mechanisms at representative clinical concentrations, with a focus on DOX behavior in breast cancer cells. It provides insight into the mechanistic pathway(s) of DOX at levels found within patients and establishes the magnitude of effect for each mechanism.

Therapy-induced polyploidization and senescence: Coincidence or interconnection?

Ewa Sikora et al.

Summary: Polyploid somatic cells play important roles in normal development and stress response. However, the transient polyploidy observed in early stages of tumors and during cancer treatment seems to be associated with tumor development and relapse. The mechanistic connection between therapy-induced senescence and polyploid formation needs to be urgently addressed, along with the question of reversibility of cancer cell senescence.

Targeting tumor cell senescence and polyploidy as potential therapeutic strategies

Tareq Saleh et al.

Summary: Cellular senescence is a complex process that can have both inhibitory and promoting effects on cancer progression. Polyploidy in tumor cells contributes to escaping growth arrest, maintaining energy production, chromatin remodeling, and drug resistance. Targeting polyploid tumor cells pharmacologically may lead to the identification of novel targets for eliminating senescent tumor cells.

Considering therapy-induced senescence as a mechanism of tumour dormancy contributing to disease recurrence

Tareq Saleh et al.

Summary: Senescence could serve as a pathway to tumour dormancy, and targeting senescent tumour cells with drugs could reduce the risk of recurrence and enhance the efficacy of cancer chemotherapy.

A common signature of cellular senescence; does it exist?

Ewa Sikora et al.

Summary: Cellular senescence is a significant biological phenomenon that mainly participates in aging and aging-related diseases through secretion activity and cell cycle arrest. In this process, DNA damage, nuclear changes, and chromatin rearrangement play important roles.

Lipid droplet evolution gives insight into polyaneuploid cancer cell lipid droplet functions

Laurie G. Kostecka et al.

Summary: Lipid droplets (LDs), originally energy stores, have evolved through different phyla to regulate lipid metabolism, signaling, and trafficking. High LD levels in cancer cells, especially in polyaneuploid cancer cells (PACC), are linked to increased tumor aggressiveness and chemotherapy resistance. PACCs are believed to be mediators of metastasis and resistance in various cancers.

Polyaneuploid Cancer Cell Dormancy: Lessons From Evolutionary Phyla

Laurie G. Kostecka et al.

Summary: Dormancy is a crucial survival strategy for many organisms facing adversity, allowing them to conserve energy by reducing metabolic rates and physical activity. Once environmental conditions improve, organisms can emerge from dormancy and readjust to their surroundings.
