Nuclear sensing of breaks in mitochondrial DNA enhances immune surveillance

Marco Tigano et al.

Summary: The study reveals that mtDSBs activate a type-I interferon response, leading to the phosphorylation of STAT1 and activation of interferon-stimulated genes. After breaks in mtDNA, herniation mediated by BAX and BAK releases mitochondrial RNA into the cytoplasm and triggers a RIG-I-MAVS-dependent immune response.

Novel Strategies for Disrupting Cancer-Cell Functions with Mitochondria-Targeted Antitumor Drug-Loaded Nanoformulations

Khaled S. Allemailem et al.

Summary: The dysregulation of mitochondria plays a significant role in cancer initiation and progression. Targeting mitochondria with therapeutic drugs using nanotechnology presents a promising approach for cancer treatment, but it is a challenging task due to multiple barriers. Novel mitochondria-selective compounds and approved nanoformulations show potential as effective anticancer agents but further research is needed to fully understand their mechanisms and optimize their use in clinical applications.

Toxicity of gold nanoparticles (AuNPs): A review

A. Sani et al.

Summary: Gold nanoparticles have great potential in biomedicine but may also have toxicity, primarily affecting the liver, spleen, and kidneys. Studies show controversy, but toxicity is mainly determined by the properties and preparations of NPs, with functionalizing NP surfaces potentially regulating and detoxifying their uptake.

Protocol to profile the bioenergetics of organoids using Seahorse

Marlies C. Ludikhuize et al.

Summary: Addressing bioenergetics is crucial for evaluating the impact of metabolism on the regulation of biological processes and its alteration in disease. Organoids are in vitro self-organizing structures derived from healthy and diseased tissues that recapitulate tissue of origin accurately, but applying Seahorse XF analysis to organoid studies poses technical challenges.
