Ankylosing Spondylitis Patients Display Aberrant ERAP1 Gene DNA Methylation and Expression

Summary: Our study found that ERAP1 gene is significantly hypermethylated and mRNA levels of EARP1 decreased in AS patients. The differential methylated CpG loci of ERAP1 showed high sensitivity, specificity, and area under curve for AS diagnosis. Furthermore, the methylation levels of ERAP1 were associated with family history, NSAID use, X-ray classification, and clinical manifestations in AS patients.

B7-H3 regulates osteoclast differentiation via type I interferon-dependent IDO induction

Summary: B7-H3 has been found to be highly expressed in mature osteoclasts, and deficiency of B7-H3 inhibits osteoclastogenesis in human osteoclast precursors by upregulating IFN signaling and IFN-inducible genes. Pharmacological inhibition and knockdown experiments can reverse the B7-H3 deficiency-mediated suppression of osteoclastogenesis.

Summary: Hypermethylation of the TRAF5 gene may contribute to the pathogenesis of AS, as the CpG island of the TRAF5 gene promoter was highly methylated in AS patients, and the relative mRNA expression level of TRAF5 was significantly reduced. Additionally, there was a negative correlation between the methylation level and mRNA level of the TRAF5 gene in AS patients.

Summary: This study found that the genetic variation of B7-H4 gene, specifically the rs10801935 and rs3738414 polymorphisms, may be correlated with susceptibility to ankylosing spondylitis (AS), particularly in male patients. The haplotype block TAG of B7-H4 was identified as a risk factor for the onset of AS in the eastern Chinese Han population. However, the association did not reach statistical significance after Bonferroni correction.
