Bariatric surgery reveals a gut-restricted TGR5 agonist with anti-diabetic effects

Snehal N. Chaudhari et al.

Summary: After bariatric surgery, levels of the endogenous bile acid cholic acid-7-sulfate (CA7S) increase in the gastrointestinal tract of both mice and humans. CA7S acts as a G-protein-coupled receptor TGR5 agonist to increase glucose tolerance during insulin resistance. This agonist remains gut-restricted, minimizing off-target effects previously observed for TGR5 agonists absorbed into the circulation.

MOLECULAR PHYSIOLOGY OF BILE ACID SIGNALING IN HEALTH, DISEASE, AND AGING

Alessia Perino et al.

Summary: Bile acids have become important signaling molecules in the body, enabling inter-tissue communication and exerting pleiotropic physiological effects. The chemical variety of bile acids, largely determined by the gut microbiome, allows for complex adaptive responses in the body. This review discusses the mechanisms by which bile acid receptors coordinate various aspects of physiology and presents new therapeutic strategies for diseases related to pathological bile acid signaling.

Hyocholic acid species improve glucose homeostasis through a distinct TGR5 and FXR signaling mechanism

Xiaojiao Zheng et al.

Summary: This study reveals that high levels of hyocholic acid (HCA) in pigs contribute to their resistance against type 2 diabetes, as depletion of bile acids (BA) in pigs leads to reduced glucagon-like peptide-1 (GLP-1) secretion and increased blood glucose levels. HCA administration in diabetic mouse models effectively improves GLP-1 secretion and glucose homeostasis by activating TGR5 and inhibiting FXR in enteroendocrine cells, a mechanism not seen in other BA species. Clinical findings also show a correlation between lower levels of HCA species and diabetes, indicating its importance in glycemic control.

Conjugated C-6 hydroxylated bile acids in serum relate to human metabolic health and gut Clostridia species

Anders O. Petersen et al.

Summary: Knowledge about the effects of C-6 hydroxylated bile acids circulating in humans, known as muricholic acids, is limited. This study found that these BAs are associated with markers of metabolic health and specific gut microbiome communities. Further research is needed to explore the therapeutic potential of these BAs in obesity and type 2 diabetes through preclinical interventions and human feasibility studies.

TGR5 controls bile acid composition and gallbladder function to protect the liver from bile acid overload

Valeska Bidault-Jourdainne et al.

Summary: This study demonstrates the importance of bile acid composition for liver protection in both mice and humans, with TGR5 identified as a key regulator of BA profile and a potential hepatoprotective target under BA overload conditions.
