Characterization of a patient-derived variant of GPX4 for precision therapy

Hengrui Liu et al.

Summary: A homozygous point mutation in the GPX4 gene, resulting in an R152H coding mutation, was identified in patients with Sedaghatian-type spondylometaphyseal dysplasia. The missense variant destabilized a critical loop in the enzyme, disrupting the active site and causing a loss of enzymatic function. Therapeutic treatments targeting the impaired R152H GPX4 activity were identified, including selenium supplementation, selective antioxidants, and a deuterated polyunsaturated fatty acid.

Selenium-Catalyzed Reduction of Hydroperoxides in Chemistry and Biology

Laura Orian et al.

Summary: Selenium plays a key role in catalyzing H2O2 reduction, but over-optimization of H2O2 reduction may lead to unexpected disturbances in metabolic regulation. Progress has been made in synthesizing organoselenides to mimic the biological strategy of selenoenzymes, but their therapeutic application remains limited.

Modeling the Catalytic Cycle of Glutathione Peroxidase by Nuclear Magnetic Resonance Spectroscopic Analysis of Selenocysteine Selenenic Acids

Ryosuke Masuda et al.

Summary: This study provides the first direct spectroscopic evidence for the formation of Sec-SeOH in small-molecule selenocysteine and selenopeptide model systems. The catalytic cycle of glutathione peroxidase was investigated using NMR-observable Sec-SeOH models, examining various chemical processes including intramolecular cyclization.

Going Forward and Back: The Complex Evolutionary History of the GPx

Thomaz Stumpf Trenz et al.

Summary: Glutathione peroxidases (GPxs) are essential antioxidant enzymes that originated from a common ancestor and evolved independently across different kingdoms. In Metazoa, GPx genes expanded into three main groups before the emergence of bilaterian animals, and further expanded in vertebrates, leading to structural and functional diversification. There is significant diversity among GPx enzymes in terms of function, structure, and evolutionary history, with ongoing birth-and-death processes occurring independently in different kingdoms and phyla.
