Super-resolution imaging illuminates new dynamic behaviors of cellulose synthase

Sydney G. Duncombe et al.

Summary: Confocal imaging has limitations in exploring the dynamic behaviors of CESA particles, but Structured Illumination Microscopy (SIM) provides a two-fold improvement in resolution and reveals new insights into the dense arrangement of cellulose. SIM data show coordinated control of CESA catalytic activity, variability in CESA track patterns, and highlight SIM as a powerful tool for advancing CESA imaging beyond conventional light microscopy.

Cell biology of primary cell wall synthesis in plants

Ying Gu et al.

Summary: Building a complex structure like the cell wall is a well-orchestrated process that requires assembling many individual parts from different sources within the cell. Cell wall components are actively transported to and from the plasma membrane during diffuse growth. Motor proteins and interactions with the cytoskeleton help transport polysaccharides and enzymes to the correct location for cell wall construction, while newly formed cell walls during cytokinesis have special features.

Multiple Arabidopsis galacturonosyltransferases synthesize polymeric homogalacturonan by oligosaccharide acceptor-dependent or de novo synthesis

Kristen A. Engle et al.

Summary: Homogalacturonan (HG) is a crucial pectic glycan in plant growth, development, and defense against pathogens, synthesized by members of the GALACTURONOSYLTRANSFERASE (GAUT) family. Different GAUT members exhibit varying levels of HG:GalAT activity, with GAUT13 and GAUT14 showing unique de novo HG synthesis capabilities.

Dynamic imaging of cell wall polysaccharides by metabolic click-mediated labeling of pectins in living elongating cells

Marc Ropitaux et al.

Summary: Protein tracking in living plant cells is routine, but tracking glycans is challenging due to their complex formation process. Currently, imaging of cell wall polymers relies on antibodies or dyes, which do not allow visualization of live cells. Click chemistry in plant cell wall biology offers an alternative by incorporating a chemical reporter into the target polysaccharide, allowing covalent linkage to a fluorescent probe. This method enables the imaging of cell wall polysaccharides in living and elongating cells.

A biophysical model for plant cell plate maturation based on the contribution of a spreading force

Muhammad Zaki Jawaid et al.

Summary: The late-stage onset of a spreading and stabilizing force is crucial for the development and maturation of plant cell plates. Researchers have used a model to study the process and found that a suitable spreading and stabilizing force is necessary for the formation and development of cell plates, and the absence of this force may lead to failure in cell plate maturation. Additionally, the findings suggest that callose may play a key role in establishing the spreading force.

Division site determination during asymmetric cell division in plants

Peishan Yi et al.

Summary: Plants use common strategies to establish asymmetric cell division, and have evolved unique cytoskeletal structures to tackle this problem. Membrane proteins localized at the polar region play a critical role in asymmetric division. This review provides updates on polarity establishment and nuclear positioning during asymmetric cell division in plants.

Building an extensible cell wall

Daniel J. Cosgrove

Summary: This article discusses the evolution of primary cell wall structure and the mechanism of surface enlargement, highlighting the role of cellulose, xyloglucan, and pectins. By using molecular dynamics methods, a quantitative model is developed to understand the mechanical behavior of the cell wall, revealing the dominant role of cellulose-cellulose interactions in forming a strong yet extensible network.

Imaging plant cells and organs with light-sheet and super-resolution microscopy

Miroslav Ovecka et al.

Summary: The documentation of plant growth and development requires integrative and scalable approaches. Recent developments in mesoscopy, microscopy and nanoscopy methods have been applied to imaging of plant subcellular compartments, cells, tissues, and organs. However, the current LSFM and SRM modalities have limitations and there is a trend towards designing integrated microscopic modalities for broader multiscale plant imaging.

Cycling in a crowd: Coordination of plant cell division, growth, and cell fate

Robert Sablowski et al.

Summary: Plant growth relies on the continuous production of new cells, with morphogenesis depending on the rate and direction of cell division and growth. Coordination between cell cycle progression, division orientation, growth cues, and cell fate changes is crucial. The retinoblastoma pathway and microtubule dynamics play key roles in linking cell cycle with growth, cell identity, and oriented cell divisions.

Cytokinesis in fra2 Arabidopsis thaliana p60-katanin Mutant: Defects in Cell Plate/Daughter Wall Formation

Emmanuel Panteris et al.

Summary: The malfunction of p60-katanin in cell plate/daughter wall formation in the fra2 Arabidopsis thaliana loss-of-function mutant was investigated. Deviations in the chemical composition of cell plate/new cell wall were identified by transmission electron microscopy (TEM), immunolabeling, and confocal microscopy. Delayed cytokinesis in the mutant results in a loss of synchronization between cell plate growth and chemical maturation.

Polysaccharide Biosynthesis: Glycosyltransferases and Their Complexes

Olga A. Zabotina et al.

Summary: Glycosyltransferases (GTs) are enzymes that attach activated sugar to acceptor substrates. While progress has been made in cloning genes encoding polysaccharide-synthesizing GTs, the majority of GTs remain uncharacterized. The mechanism by which they synthesize complex polysaccharide structures in the Golgi membrane is largely unknown.
