Co-aggregation and secondary nucleation in the life cycle of human prolactin/galanin functional amyloids

Summary: The synergistic aggregation and cross-seeding of two different proteins/peptides in amyloid aggregation play a crucial role in various neurological disorders, including Alzheimer's disease. In this study, the co-storage of human Prolactin (PRL) and neuropeptide galanin (GAL) in secretory granules (SGs) of female rats is demonstrated as functional amyloids. Despite differences in sequence and structure, both hormones facilitate their synergistic aggregation into amyloid fibrils. Furthermore, PRL seeds can initiate GAL aggregation, while mixed fibrils cannot facilitate cross-seeding, suggesting tight regulation of functional amyloid formation by these hormones for efficient storage in SGs. The faster release of functional hormones from mixed fibrils compared to individual amyloids suggests a novel mechanism of heterogeneous amyloid formation in functional amyloids of pituitary SGs.
