A teamwork promotion of formin-mediated actin nucleation by Bud6 and Aip5 in Saccharomyces cerevisiae

Ying Xie et al.

Summary: Actin nucleation is achieved through collaborative teamwork between actin nucleator factors (NFs) and nucleation-promoting factors (NPFs). In budding yeast, the formins Bni1 and Bnr1 team up with different NPFs, but the selective pairing between formin NFs and NPFs is not fully understood. Through the examination of NPFs and NFs functions and interactions, it was found that Aip5 and Bud6 interact with the C-terminal intrinsically disordered region (IDR) of formin, promoting formin-mediated actin nucleation.

Potentiation of plant defense by bacterial outer membrane vesicles is mediated by membrane nanodomains

Tuan Minh Tran et al.

Summary: This study demonstrates that interactions between OMVs and the Arabidopsis thaliana plasma membrane can modulate various host processes, including endocytosis, immune responses, and suppression of pathogenesis. OMVs have a highly ordered lipid phase and directly insert into the plant PM, enhancing lipid order and strengthening plant defenses; this process is dependent on host nanodomains and remorin. Computational simulations show that OMV integration into the plant PM depends on membrane lipid order and the saturation level of OMV lipids can fine-tune the enhancement of host lipid order.

Membrane nanodomains modulate formin condensation for actin remodeling in Arabidopsis innate immune responses

Zhiming Ma et al.

Summary: This study demonstrates that the condensation of nanodomain constituents and the self-assembly of remorin proteins regulate the condensation and activity of formins during innate immunity in Arabidopsis thaliana. Through interaction with formins, remorin enhances actin nucleation, influencing the transduction of immune signals. Such nanodomain- and remorin-mediated regulation is expected to be a common feature in plant innate immune responses, creating spatially separated biochemical compartments and fine-tuning membrane properties for immune signal transduction in the host.

Expression Levels of Genes Encoding Proteins Involved in the Cell Wall-Plasma Membrane-Cytoskeleton Continuum Are Associated With the Maturation-Related Adventitious Rooting Competence of Pine Stem Cuttings

Alberto Pizarro et al.

Summary: This study identified pine homologs of genes involved in the cell wall-plasma membrane-cytoskeleton continuum and analyzed their expression levels during adventitious root formation in Pinus radiata. The expression of genes involved in sensing cell wall and membrane disturbances was found to be more correlated with adventitious root formation competence than with auxin and time of induction. These findings provide insights into the recalcitrance of stem cuttings to adventitious root formation.

Salicylic acid regulatesPIN2auxin transporter hyperclustering and root gravitropic growth viaRemorin-dependent lipid nanodomain organisation inArabidopsis thaliana

Meiyu Ke et al.

Summary: Plants adapt to environmental cues through sophisticated mechanisms that involve growth-promoting hormones and defence signaling, especially the interaction between salicylic acid (SA) and auxin. The study reveals how SA regulates auxin signaling by modulating the membrane dynamics of PIN2 transporter, influencing auxin distribution and gravitropic responses. This defense-growth trade-off mechanism involves a crosstalk that concentrates PIN2 into membrane compartments, disrupting auxin transport.

Polarisome assembly mediates actin remodeling during polarized yeast and fungal growth

Ying Xie et al.

Summary: The dynamic assembly and remodeling of actin in filamentous fungi and budding yeast plays a crucial role in establishing and maintaining polarity, with the polarisome at the tip region driving actin cable polymerization. Understanding the regulated interactions within the polarisome is essential for comprehending the tunable activities of the formin-centered nucleation core. Furthermore, the distribution and mechanical feedback mechanisms of polarisome components play a significant role in regulating polarized growth.

Formin-mediated bridging of cell wall, plasma membrane, and cytoskeleton in symbiotic infections of Medicago truncatula

Pengbo Liang et al.

Summary: Legumes have the ability to associate with rhizobia for nitrogen-fixing root nodule symbiosis. In Medicago truncatula, root hair colonization by Sinorhizobium meliloti begins from young root hairs, which physically entwine around the symbiont to form rhizobial traps. This unique process involves alterations in membrane organization, cytosolic calcium gradient, actin rearrangements, and symbiotic responses in legumes.
