The impact of protein corona on the biological behavior of targeting nanomedicines

Summary: In order to successfully translate targeting nanomedicines from bench to bedside, it is necessary to address the common issues of rapid clearance and off-target accumulation. This phenomenon, known as protein corona (PC) formation, occurs when targeting nanoparticles encounter the bloodstream and interact with plasma proteins. The present review discusses the impact of PC on the biological behavior of targeting nanomedicines, as well as the best simulated environment and latest characterization techniques for PC analysis. The article also highlights recent progress in manipulating nano-bio interfaces to achieve optimal therapeutic outcomes.

Comparison of three synthetic transferrin mimetic small peptides to promote the blood-brain barrier penetration of vincristine liposomes for improved glioma targeted therapy

Summary: The development of a modified liposomal formulation has shown promising results in facilitating brain targeted delivery and selective accumulation within glioma cells. Among the studied targeting ligands, T7-modified liposomes demonstrated superior BBB penetration, brain distribution, and increased accumulation in glioma cells, leading to improved anti-glioma outcomes through targeted cytotoxicity and apoptosis. These results suggest that T7-modified liposomes could serve as a potential platform for effective brain targeted delivery and glioma therapy in clinical settings.

Research has shown promising results with nanoparticles (NPs-K-s-A) targeting LRP1 and MMP1 to overcome the blood-brain barrier and enhance drug delivery, leading to improved therapeutic outcomes in mice with BCBMs. This fusion peptide-based drug delivery strategy shows potential for clinical management of BCBMs.

Transferrin-functionalized liposomes for docetaxel delivery to prostate cancer cells

Summary: This study investigated the delivery of docetaxel to prostate cancer cells using transferrin-functionalized liposomes. Results showed that these systems had small and stable size, with higher toxicity towards PC-3 cells. Confocal microscopy confirmed successful internalization of functionalized liposomes in PC-3 cells, while interaction with transferrin receptors was further validated using Quartz Crystal Microbalance with Dissipation technology. Further studies are needed to explore the potential of these systems in three-dimensional cell models and in vivo xenograft studies.
