Summary: Vulvar squamous cell carcinomas (VSCC) can be classified into HPV(+) and HPV(-) subgroups, with significant molecular differences between the two. TP53 mutations are the most common somatic mutations in VSCC, with CDKN2A mutations more common in HPV(-) VSCC, and the PTEN/PI3K/AKT/mTOR pathway commonly altered in HPV(+) VSCC, potentially providing targetable options.
