Characterization of Opposing Responses to Phenol by Bacillus subtilis Chemoreceptors

Girija A. Bodhankar et al.

Summary: This study investigates the chemotaxis of Bacillus subtilis to phenol. The main findings are that McpA is the principal chemoreceptor responsible for the repellent response to phenol, while McpC and HemAT govern the attractant response to phenol. The signaling domain of McpA is found to sense phenol, as evidenced by in vitro binding experiments. These results contribute to our understanding of bacterial chemotaxis and demonstrate the ability of chemoreceptors to directly sense chemoeffectors.

A Dual Regulatory Role of the PhoU Protein in Salmonella Typhimurium

Soomin Choi et al.

Summary: Bacteria use two-component systems to sense and respond to their environment. The PhoU protein, along with other components, plays a role in phosphate signaling in Gram-negative bacteria. PhoU is believed to function as a negative regulator in phosphate signaling, but its exact mechanism is not fully understood. This study identified specific residues in PhoU that interact with the PhoR histidine kinase and found that these interactions control the expression of pho genes in different phosphate and Mg2+ concentrations. Furthermore, it was discovered that PhoU's role in promoting PhoR autophosphorylation is required for Salmonella virulence.

A catalogue of signal molecules that interact with sensor kinases, chemoreceptors and transcriptional regulators

Miguel A. Matilla et al.

Summary: Bacteria have developed various signal transduction systems to sense and respond to signals, with transcriptional regulators, sensor histidine kinases, and chemoreceptors being the most prevalent. These systems use ligand-binding domains (LBDs) to recognize signal molecules and modulate the activity of the output module. By manually retrieving information from publications and 3D structures, the authors classified 811 proteins, revealing the signal profiles and distinguishing between families with promiscuous signal recognition and those with well-defined ligand ranges. This signal repertoire will aid in identifying signaling molecules for uncharacterised sensor proteins.

Conformation control of the histidine kinase BceS ofBacillus subtilisby its cognate ABC-transporter facilitates need-based activation of antibiotic resistance

Alan Koh et al.

Summary: Bacteria tightly control gene expression to minimize fitness costs associated with antibiotic resistance. In Bacillus subtilis, a novel regulatory logic involving a two-component system and an ABC transporter allows direct monitoring of detoxification needs. The transporter not only activates the kinase, but also helps maintain its inactive state, ensuring precise flux-dependent signaling control. Transport flux-dependent signaling conserves energy and provides a unique strategy for energy-efficient decision making.

PAS domains in bacterial signal transduction

Erwin C. Stuffle et al.

Summary: PAS domains are versatile domains found in proteins across different kingdoms, with a conserved cleft for cofactor or ligand binding. Recent years have seen a significant increase in identified PAS domains and corresponding structures, leading to a better understanding of PAS signaling mechanisms. New bacterial PAS ligands have been discovered, expanding the list of bacterial PAS functions beyond signal sensing to include modulation, transduction, dimerization, protein interaction, and cellular localization.
