Large-scale GWAS reveals genetic architecture of brain white matter microstructure and genetic overlap with cognitive and mental health traits (n=17,706)

Bingxin Zhao et al.

Summary: The study analyzed the genetic architecture of 110 tract-based DTI parameters and conducted GWAS to identify novel genetic variants associated with white matter tracts. Results suggested a highly polygenic architecture of genetic influence across the genome, with enrichment of genetic effects for active SNPs annotated by central nervous system cells. GWAS identified 213 significant SNPs associated with 90 DTI parameters, and gene-based association study prioritized 112 significant genes, many of which are novel and have implications for cognitive and mental health traits.

Metformin Benefits: Another Example for Alternative Energy Substrate Mechanism?

Andrea Giaccari et al.

Summary: Metformin, considered the first-line medication for newly diagnosed type 2 diabetes patients, may protect from diabetic and non-diabetic comorbidities by increasing lactate production and oxidation, thereby potentially exerting a protective effect on the heart, kidneys, brain, and antineoplastic action.

Effects of linagliptin vs glimepiride on cognitive performance in type 2 diabetes: results of the randomised double-blind, active-controlled CAROLINA-COGNITION study

Geert Jan Biessels et al.

Summary: The study assessed the impact of the DPP-4 inhibitor linagliptin compared to the sulfonylurea glimepiride on accelerated cognitive decline in individuals with type 2 diabetes. Results showed no significant difference in the risk for cognitive decline between the two drugs over a 6.1-year period in people with early type 2 diabetes at elevated cardiovascular risk.

Safety and efficacy of pioglitazone for the delay of cognitive impairment in people at risk of Alzheimer's disease (TOMMORROW): a prognostic biomarker study and a phase 3, randomised, double-blind, placebo-controlled trial

Daniel K. Burns et al.

Summary: The study aimed to identify participants at risk of developing mild cognitive impairment due to Alzheimer's disease using the BRAA algorithm and evaluate the safety and efficacy of low-dose pioglitazone. Results showed that pioglitazone did not delay the onset of mild cognitive impairment.

The trans-ancestral genomic architecture of glycemic traits

Ji Chen et al.

Summary: A meta-analysis of GWAS including up to 281,416 individuals without diabetes identified 242 loci associated with glycemic traits, with 99 novel loci discovered. The trans-ancestry approach in the study improved the discovery of new loci and the fine-mapping of variants.

Large-scale cis- and trans-eQTL analyses identify thousands of genetic loci and polygenic scores that regulate blood gene expression

Urmo Vosa et al.

Summary: Genetic variants associated with traits primarily affect complex phenotypes through regulatory mechanisms on the transcriptome. Most genes show cis-eQTL effects, while distal trans-eQTL effects have lower replication rates. However, single-cell RNA-seq data prioritize intracellular trans-eQTL effects mediated by transcription factors. Additionally, a subset of genes correlated with polygenic scores for various phenotypes may serve as potential drivers for those traits.
