Hong Sun et al.

Summary: This study provides global, regional, and country-level estimates of diabetes prevalence and health expenditures for 2021 and projections for 2045. The global prevalence of diabetes is increasing and is expected to have the greatest relative increase in middle-income countries. Diabetes-related health expenditures are also projected to rise.

Coronary Heart Disease in Type 2 Diabetes Mellitus: Genetic Factors and Their Mechanisms, Gene-Gene, and Gene-Environment Interactions in the Asian Populations

Khairul Anwar Zarkasi et al.

Summary: Asians are more susceptible to type 2 diabetes and coronary heart disease complications compared to Western populations, possibly due to genetic factors, obesity, insulin resistance, and endothelial dysfunction. Genetic factors play a significant role in energy and lipoprotein metabolism, vascular and endothelial pathology, antioxidation, and gene-environment interactions can modify the disease risk. These factors highlight the complexities of coronary heart disease in type 2 diabetes Asian populations.

Summary: The study revealed a close association between FPG levels and cardiovascular disease risk in young adults, with IFG and DM increasing the risk of heart attack, angina pectoris, stroke, heart failure, and atrial fibrillation. The findings emphasize the importance of maintaining optimal FPG levels for primary prevention of CVD in young adults.

Summary: Patients with chronic kidney disease face an increased risk of cardiovascular events, with advanced CKD stages significantly elevating the risk. Cardiovascular events, rather than end-stage kidney disease, are the leading cause of death in this high-risk population, and CKD causes a proinflammatory state that affects the cardiovascular system.

Cardiac Energy Metabolism in Heart Failure

Gary D. Lopaschuk et al.

Summary: Changes in cardiac energy metabolism are complex and depend on the severity and type of heart failure, as well as the presence of common comorbidities such as obesity and type 2 diabetes. The failing heart experiences an energy deficit due to decreased mitochondrial oxidative capacity, compensated for by increased ATP production from glycolysis. Alterations in mitochondrial ATP production fuels and fatty acid oxidation contribute to the inefficiency of the failing heart, with potential therapeutic targets in energy metabolic pathways to improve cardiac function.

Structural basis of ethnic-specific variants of PAX4 associated with type 2 diabetes

Summary: By conducting a genome-wide association study of type 2 diabetes in a Japanese population, we identified 20 novel T2D loci that were not associated with T2D in Europeans, as well as nine novel missense risk variants, such as those of PAX4, which were common in the Japanese population but rare in Europeans. Our in silico structural analysis of ethnic-specific variants of PAX4 suggests their pathogenic effect.
