Summary: The study found that even minimal residual C-peptide secretion could have clinical benefits in type 1 diabetes. The effects on the risk of hypoglycemic episodes were detectable at lower levels of C-peptide.

Efficacy and safety of adding sotagliflozin, a dual sodium-glucose co-transporter (SGLT)1 and SGLT2 inhibitor, to optimized insulin therapy in adults with type 1 diabetes and baseline body mass index ≥ 27 kg/m2

Thomas Danne et al.

Summary: Sotagliflozin, as an adjunct to optimized insulin therapy in overweight/obese patients with type 1 diabetes, demonstrates significant efficacy and safety in patients with BMI >= 27 kg/m(2). It reduces body weight, lowers systolic blood pressure, helps achieve optimal glycaemic control, and does not increase the risk of hypoglycaemia.

The Impact of Low-dose Gliclazide on the Incretin Effect and Indices of Beta-cell Function

Summary: The study showed that low-dose gliclazide reduces blood glucose levels in T2DM patients, while enhancing the classical incretin effect. This medication can increase beta-cell glucose sensitivity and potentiate late-phase incretin action.

The Role of Incretins on Insulin Function and Glucose Homeostasis

Summary: The incretin effect refers to the increase in insulin secretion after oral and intravenous glucose administration, mediated by the hormones GIP and GLP-1. In healthy individuals, GIP contributes the most to postprandial insulin secretion. The loss of incretin effect in diabetes significantly affects postprandial blood sugar control.

Reappearance of C-Peptide During the Third Trimester of Pregnancy in Type 1 Diabetes: Pancreatic Regeneration or Fetal Hyperinsulinism?

Claire L. Meek et al.

Summary: Three distinct patterns of maternal C-peptide trajectory were observed in pregnant women with type 1 diabetes, with the appearance of C-peptide at 34 weeks associated with midtrimester hyperglycemia, elevated cord blood C-peptide, and high rates of neonatal complications such as large for gestational age, neonatal hypoglycemia, and neonatal intensive care admission. This finding suggests the transfer of C-peptide from fetal to maternal serum and is inconsistent with the hypothesis of pregnancy-related beta-cell regeneration.

Latent Autoimmune Diabetes of Adults (LADA) Is Likely to Represent a Mixed Population of Autoimmune (Type 1) and Nonautoimmune (Type 2) Diabetes

Angus G. Jones et al.

Summary: LADA, diagnosed after age 35, presents clinical features similar to type 2 diabetes but with type 1 diabetes-associated islet autoantibodies. The current definition of LADA identifies a group with features intermediate between typical type 1 and type 2 diabetes, leading to difficulties in classification. Improved approaches are needed to increase test specificity and better understand the detailed pathophysiological components of late-onset autoimmune diabetes.

Proinsulin:C-peptide ratio trajectories over time in relatives at increased risk of progression to type 1 diabetes

Taylor M. Triolo et al.

Summary: In cross-sectional analyses, there were no significant differences in PI:C ratios between relatives with different levels of autoantibodies. However, over time, the trajectories of PI:C ratios increased in individuals with multiple autoantibodies or those who progressed to type 1 diabetes, suggesting it may be a useful biomarker for assessing the risk of progression to type 1 diabetes.

The Role of Incretins on Insulin Function and Glucose Homeostasis
