Beneficial effects of citrulline enteral administration on sepsis-induced T cell mitochondrial dysfunction

Florian Reizine et al.

Summary: Severe sepsis can lead to sustained immune dysfunction, resulting in organ failure, secondary infections, and late mortality. Supplementation with arginine or citrulline can restore T cell function and reduce immunosuppression induced by sepsis.

Inhibiting KDM6A Demethylase Represses Long Non-Coding RNA Hotairm1 Transcription in MDSC During Sepsis

Isatou Bah et al.

Summary: Myeloid-derived suppressor cells (MDSCs) prolong sepsis by promoting immunosuppression. The development of sepsis MDSCs requires the interaction between long non-coding RNA Hotairm1 and S100A9. Histone demethylase KDM6A promotes Hotairm1 transcription by demethylating transcription repression H3K27me3 histone mark. Both IL-10 cytokine and GSK-J4 treatment can affect Hotairm1 transcription.

SARS-CoV-2-Induced ARDS Associates with MDSC Expansion, Lymphocyte Dysfunction, and Arginine Shortage

Florian Reizine et al.

Summary: The study revealed that ARDS patients with COVID-19 had increased MDSC and decreased CD8(pos) effector memory cell compared to patients with moderate COVID-19 pneumonia. The expansion of COVID-19-related MDSC was directly correlated to lymphopenia and enhanced arginase activity. T cell proliferative capacity in vitro was significantly reduced among COVID-19 patients but could be restored through arginine supplementation.

GM-CSF Nitration Is a New Driver of Myeloid Suppressor Cell Activity in Tumors

Bianca Cali et al.

Summary: Reactive oxygen species, including RNS, play a crucial role in controlling immune cell functions within the tumor microenvironment, and GM-CSF activity is significantly impacted by RNS-induced post-translational modifications. The nitration of a single tryptophan residue in GM-CSF sequence facilitates the expansion of highly immunosuppressive myeloid subsets in tumor-bearing hosts, providing a potential target for remodeling the TME and enhancing protective immunity against cancer.

Isoform specific anti-TGFβ therapy enhances antitumor efficacy in mouse models of cancer

Aditi Gupta et al.

Summary: The study highlights the potential of TGF beta as a target in cancer treatment, showing that myeloid/dendritic cells are main sources of TGF beta 1 and TGF beta 3 in stroma-poor tumors. Targeting isoform-specific TGF beta leads to enhanced CD8 T cell function and improved tumor control when compared to pan-TGF beta inhibition. Combining TGF beta inhibition with immune checkpoint blockade results in a more effective tumor control by shifting the local immune environment to favor tumor suppression.

Liver X receptor agonist GW3965 protects against sepsis by promoting myeloid derived suppressor cells apoptosis in mice

Wenqin Zhang et al.

Summary: The study demonstrates that the LXR agonist GW3965 improves survival in septic mice by reducing multi-organ injury and inflammatory cytokine levels, as well as decreasing MDSC abundance in spleen and enhancing bacteria clearance in tissues. GW3965 activates LXR beta and ABCA1 to induce apoptosis of spleen MDSCs, offering a potential approach to target sepsis-induced immunosuppression.

All-trans-retinoic acid restores CD4+T cell response after sepsis by inhibiting the expansion and activation of myeloid-derived suppressor cells

Tao Liu et al.

Summary: ATRA can reduce the frequency and activity of MDSCs in the spleen of mice by regulating the expression of arginase-1 and iNOS, as well as the secretion of immune-suppressive cytokines. The administration of ATRA ultimately decreased mortality in CLP-surviving mice with secondary infection, potentially due to the restoration of CD4+ T cell function.
