Dietary Pterostilbene for MTA1-Targeted Interception in High-Risk Premalignant Prostate Cancer

Summary: This study investigates the role of MTA1 and PTEN deficiency in the development of prostate cancer and the chemopreventive effect of pterostilbene. The results reveal that MTA1 promotes neoplastic transformation of prostate epithelial cells and cooperates with PTEN deficiency to accelerate PIN development. Furthermore, a diet supplemented with pterostilbene reduces the severity and number of PIN foci and decreases cell proliferation, angiogenesis, and inflammation. Therefore, pterostilbene shows promise as a chemopreventive agent for inhibiting cancer development in active surveillance patients with prostate cancer.

MTA1, a metastasis-associated protein, in endothelial cells is an essential molecule for angiogenesis

Summary: Our study demonstrated the importance of MTA1 expression in tube formation. Knockout of MTA1 inhibited tube formation, while overexpression of MTA1 restored the ability to form tube-like structures. Furthermore, MTA1 knockout cells showed increased phosphorylation of non-muscle myosin heavy chain IIa, leading to the suppression of tube formation. These findings suggest that MTA1 may play a crucial role in angiogenesis in endothelial cells.

Summary: The study revealed that treatment with CN133 resulted in a 50% reduction in tumor volume, which was more effective than SAHA treatment. [C-11] Martinostat PET imaging was able to identify early lesions of prostate cancer and monitor the therapeutic effect of CN133 in CRPC. Pharmacological approaches demonstrated that CN133 showed nearly 100-fold efficacy compared to SAHA treatment in cell proliferation, invasion, and migration experiments.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Metastasis-associated gene 1 (MTA1) enhances cisplatin resistance of malignant pleural mesothelioma by ATR-Chk1-mediated DNA repair

Summary: In malignant pleural mesothelioma, the aberrant expression of MTA1 is associated with cisplatin resistance, with cisplatin stabilizing MTA1 expression by inhibiting ubiquitination. MTA1 enhances G2/M cell cycle delay and protects the tumor genome through participating in the ATR-Chk1 pathway. These findings suggest MTA1 could be a novel therapeutic target to overcome chemoresistance in MPM.
