The genome of flavivirus encodes ten proteins, each playing a different and crucial role in viral infection. While significant progress has been made in the understanding of flavivirus biology in recent years, there is currently a lack of fully effective antiviral drugs or vaccines for most flaviviruses.

A pan-serotype dengue virus inhibitor targeting the NS3-NS4B interaction

Summary: JNJ-A07 is a highly potent inhibitor of dengue virus, showing nanomolar to picomolar activity against a panel of 21 clinical isolates. It prevents the formation of viral replication complex by blocking the interaction between NS3 and NS4B proteins, revealing a previously undescribed mechanism of antiviral action. The molecule has a favorable pharmacokinetic profile and exhibits outstanding efficacy in mouse infection models, even when treatment is initiated at peak viraemia.

Mechanism studies suggested a suppression of viral infection during later stages of the replication cycle, with an observed cell-dependent antiviral profile against multiple Zika strains indicating potential involvement of cellular factors in the compounds' activity.

Ebselen and Analogues: Pharmacological Properties and Synthetic Strategies for Their Preparation

Summary: Ebselen, the leader of selenorganic compounds, has been identified as a mimetic of the key antioxidant enzyme glutathione peroxidase and is currently undergoing clinical evaluation for treating COVID-19 patients. Despite its lack of specificity in vitro, Ebselen has shown good tolerance in vivo with no signs of toxicity at administered doses. Additionally, there is a lack of review articles on synthetic approaches for the construction of the benzo[d][1,2]-selenazol-3(2H)-one scaffold, a topic which is addressed in this review.
