Metallaphotoredox: The Merger of Photoredox and Transition Metal Catalysis

Amy Y. Chan et al.

Summary: The merger of photoredox catalysis with transition metal catalysis, known as metallaphotoredox catalysis, has become an important part of synthetic methodology. It combines the bond formation capability of transition metal catalysis with the utility of photoinduced electron and energy transfer processes. Photocatalytic substrate activation allows the involvement of simple starting materials in metal-mediated bond-forming processes, while electron or energy transfer with organometallic intermediates provides a complementary activation mode to traditional catalytic platforms.

Development of a high intensity parallel photoreactor for high throughput screening

Ning Qi et al.

Summary: The development of a high intensity photoreactor allows for high throughput screening of up to 1536 reactions on nanomole scales in parallel, offering increased photon flux for reactions sensitive to light intensity. The reactor's uniform illumination and efficient heat removal enable systematic investigation of parameter interplay on nanomole scale, providing a standardized platform for screening and data gathering. Additionally, the reactor allows for successful reproduction of screening hits at medicinal chemistry relevant scales.

Rerouting the Organocatalytic Benzoin Reaction toward Aldehyde Deuteration

Santosh C. Gadekar et al.

Summary: Reactive intermediates play a crucial role in promoting or halting chemical reactions, but their elusive nature often makes them challenging to harness directly for reaction design. This study successfully stabilized reactive intermediates in the N-heterocyclic carbene (NHC) catalytic cycle, leading to the shutdown of the benzoin coupling pathway and rerouting the intermediates towards deuteration. By introducing catalytic amounts of phenyl boronic acid, highly reactive intermediates were further stabilized, mitigating the formation of benzoin coupling byproducts and allowing for a scalable deuteration reaction with outstanding functional group tolerance.

Access to Deuterated Unnatural α-Amino Acids and Peptides by Photochemical Acyl Radical Addition

Li Liu et al.

Summary: A visible-light-enabled, photocatalyst-free conjugate addition reaction of dehydroamino acids using 4-acyl-1,4-dihydropyridines as both a radical reservoir and reductant has been successfully developed. Various beta-acyl alpha-amino acids and their deuterated analogues were obtained in good results. This method features mild conditions and efficient derivatization, providing a new pathway for the synthesis of unnatural amino acid derivatives.

Acyl fluorides as direct precursors to fluoride ketyl radicals: reductive deuteration using SmI2 and D2O

Hengzhao Li et al.

Summary: A highly chemoselective reductive deuteration of acyl fluorides using SmI2 and D2O as the deuterium source has been developed, providing alpha,alpha-dideuterio alcohols with exceptional levels of deuterium incorporation. This method introduces acyl fluorides as attractive radical precursors for the generation of reactive acyl-type fluoride ketyls, which should have widespread application in synthetic strategies involving single electron transfer processes.
