Single-chain variable fragment antibody-based ic-ELISA for rapid detection of macrolides in porcine muscle and computational simulation of its interaction mechanism

Summary: The study generated a single-chain fragment variable (scFv) for the detection of macrolide antibiotics (MAs) residues through biopanning and comparative analysis, revealing that mutations in the FR framework region can affect the active pocket and recognition ability of the antibody. Through homology modeling and molecular docking, the binding sites between the optimal MA-specific scFv candidate and MA were identified.

Summary: A chemiluminescent enzyme immunoassay (CLEIA) based on a single-chain variable fragment (scFv)-alkaline phosphatase (AP) fusion protein was developed to determine malachite green (MG) and its major metabolite, leucomalachite green (LMG) residual levels in tilapia fish. The validation results of this novel competitive CLEIA were consistent with those obtained by classical HPLC method for determination of total MG in spiked and field incurred samples.

Soluble Cytoplasmic Expression and Purification of Immunotoxin HER2(scFv)-PE24B as a Maltose Binding Protein Fusion

Summary: In this study, a recombinant immunotoxin gene was successfully expressed in E. coli by fusion with MBP, resulting in soluble expression and purification of stable HER2(scFv)-PE24B. The purified HER2(scFv)-PE24B exhibited strong cytotoxic effects against four different breast cancer cell lines, with higher efficacy in high HER2-positive cell lines SKBR3 and BT-474.

Identification of chicken-derived scFv against N-glycolylneuraminic acid retrieved from an immune library by phage display

Summary: N-glycolylneuraminic acid (Neu5Gc) can cause chronic inflammation in the human body, promoting the development of cancer and related diseases. In this study, a gene sequence for Neu5Gc-specific ScFv was identified for the development of IC-ELISA and test strip to detect Neu5Gc in red meat, milk, and tumor tissues. The results showed promising potential for the specific anti-Neu5Gc ScFv as a detection probe.

Preparation and Directed Evolution of Anti-Ciprofloxacin ScFv for Immunoassay in Animal-Derived Food

The results indicate the potential for using this method in detecting CIP residues in animal-derived food products.
