Designed Antitumor Peptide for Targeted siRNA Delivery into Cancer Spheroids

Summary: The amphiphilic alpha-helical peptide G3 shows high specificity and affinity for cancer cells, making it a promising therapeutic agent for cancer treatment. The peptide enters cancer cells through an energy-dependent endocytic pathway and can protect and selectively deliver siRNAs to modulate gene expression. In vivo toxicity studies on zebrafish embryos show that G3's toxicity is concentration-dependent but can be attenuated when complexed with siRNA, making it a potential effective gene delivery agent with low toxicity.

The evolution of commercial drug delivery technologies

Three drug delivery paradigms have been developed to aid in the successful delivery of small molecules and macromolecules, contributing to the initial clinical success of each therapeutic class.

The stiffness of the nanocarriers significantly impacted their cellular uptake and anticancer efficacy, making them promising for cancer therapy.
