How to improve the efficiency of biocatalysis in non-aqueous pure deep eutectic solvents: A case study on the lipase-catalyzed transesterification reaction

Jun Cao et al.

Summary: Non-aqueous biocatalysis is expanding its application fields and playing an important role in industrial biocatalysis. Deep eutectic solvents (DESs) are recognized as green reaction media for non-aqueous biocatalysis. By studying the catalytic efficiency of Candida antarctica lipase B (CALB) in different DESs, it was found that the properties of the DESs strongly influenced CALB's catalytic efficiency. Learning from CALB's behavior in DESs, a preliminary attempt in designing more efficient DESs was provided.

Acceleration of lactose hydrolysis using beta-galactosidase and deep eutectic solvents

Jakub Hoppe et al.

Summary: A series of new deep eutectic solvents were prepared and characterized. Their physicochemical properties were determined and their effectiveness as additives in enzymatic reactions was investigated. The results showed that these deep eutectic solvents had desirable viscosity, thermal stability, and density, and they could stabilize enzymes and enhance their activity. One of the deep eutectic solvents significantly increased the lactose hydrolysis yield by more than threefold.

Enzymatic Synthesis and Molecular Modelling Studies of Rhamnose Esters Using Lipase from Pseudomonas stutzeri

Cecilia Garcia-Oliva et al.

Summary: This study reports the enzymatic synthesis of rhamnose fatty acid esters through transesterification using lipase from Pseudomonas stutzeri as a biocatalyst. Optimum conditions were determined for achieving high conversion and regioselectivity, and the role of various reaction conditions was investigated. The use of specific solvents and acyl donors with different chain lengths greatly influenced the efficiency of the reaction. The enzyme's regioselectivity and its ability to accommodate ester chains of different lengths were explained through molecular modeling and simulation studies.

Deep Eutectic Solvents: A Review of Fundamentals and Applications

Benworth B. Hansen et al.

Summary: Deep eutectic solvents (DESs) exhibit significant depressions in melting points and tunable physicochemical properties, but there is still a lack of predictive understanding of their microscopic mechanisms and structure-property relationships. Research on complex hydrogen bonding is crucial for a deeper understanding and development of fundamental frameworks for DESs.

Aqueous ionic liquids redistribute local enzyme stability via long-range perturbation pathways

Till El Harrar et al.

Summary: This study investigates the impact of aqueous ionic liquids on a model enzyme through molecular dynamics simulations and finds that they can affect enzyme activity by modifying local structural stability, eventually leading to perturbations in the catalytic site and protein core. The identification of perturbation pathways and specific interactions with IL ions helps predict variant libraries with improved tolerance to aqueous ionic liquids.
