Postweaning positive modulation of α5GABAA receptors improves autism-like features in prenatal valproate rat model in a sex-specific manner

Summary: This study investigated the effectiveness of MP-III-022, a selective positive allosteric modulator of α5GABAA receptors, in a rat model of autism. The results showed that MP-III-022 improved social, repetitive, and restrictive impairments in rats prenatally exposed to valproic acid. Interestingly, MP-III-022 also induced autism-like changes in control rats. Calcium imaging and gene expression analysis further supported the role of α5GABAA receptors in the pathophysiology of autism spectrum disorder.
