Inflammation and Immune Response in Arrhythmogenic Cardiomyopathy: State-of-the-Art Review

Babken Asatryan et al.

Summary: Arrhythmogenic cardiomyopathy (ACM) is primarily caused by genetic defects in proteins of the cardiac intercalated disc, with wide phenotype variability. Inflammation and immune responses are considered aggravators of cardiac myocyte damage, and play a role in the pathogenesis of ACM.

Exome-wide evaluation of rare coding variants using electronic health records identifies new gene-phenotype associations

Joseph Park et al.

Summary: The study examined the phenotypic effects of rare loss-of-function gene variants using 10,900 whole-exome sequences linked to electronic health records in the Penn Medicine Biobank, identifying new gene-disease associations that replicated across other biobanks.

Promise and Peril of Population Genomics for the Development of Genome-First Approaches in Mendelian Cardiovascular Disease

Victoria N. Parikh

Summary: The rich tradition of cardiovascular genomics has positioned the field to advance towards a genome-first approach, leveraging population-scale genomic data to elucidate variant pathogenicity. However, the implementation of this approach is limited by characteristics of the studied populations and evolutionary constraints on human Mendelian variation. Successful adoption of a genome-first approach to rare cardiovascular disease will require a nuanced integration of population-based discovery with detailed investigation of rare disease cohorts and prospective variant evaluation.

Evidence-Based Assessment of Genes in Dilated Cardiomyopathy

Elizabeth Jordan et al.

Summary: A systematic curation of 51 genes related to DCM revealed that 19 genes have high evidence (12 definitive/strong, 7 moderate), but these genes only explain a minority of cases, leaving the remainder of DCM genetic architecture incompletely addressed.

The Complex and Diverse Genetic Architecture of Dilated Cardiomyopathy

Ray E. Hershberger et al.

Summary: Insights into the genetic architecture of dilated cardiomyopathy (DCM) are rapidly evolving, showing a complex and diverse nature. In addition to monogenic structures, oligogenic basis, rare variants, low-frequency, and common genetic variations may contribute to the complexity of DCM, along with gene-environment interactions.
