In vitro and in vivo evaluation of 211At-labeled fibroblast activation protein inhibitor for glioma treatment

Summary: This study investigated the therapeutic effect of At-211-labeled fibroblast activation protein inhibitor (FAPI) for glioma treatment. The results showed that At-211-FAPI-04 had fast and specific binding to FAP-positive U87MG cells and significantly reduced cell viability, arrested cell cycle, and suppressed cell proliferative efficacy. Intratumoral injection of At-211-FAPI-04 resulted in 6-fold higher accumulation in tumor sites compared to intravenous injection. At-211-FAPI-04 effectively suppressed tumor growth and prolonged median survival in U87MG xenografts without obvious toxicity to normal organs. These findings suggest that targeted alpha-particle therapy mediated by At-211-FAPI-04 is a promising strategy for glioma treatment.

Both closed shell (ionic, long range) and intermediate character interactions contribute to stabilizing the clusters, with the ability of water molecules to donate or accept electron density dictating the nature and strength of hydrogen bonds in solvation shells. Binding energies and molecular geometries are shown to be more influenced by electron correlation rather than relativistic effects.

Synthesis and preliminary evaluation of 211At-labeled inhibitors of prostate-specific membrane antigen for targeted alpha particle therapy of prostate cancer

By performing a singular value decomposition, the method shows how localized valence virtual orbitals can be expressed on an intrinsic minimal basis. The developed scheme is implemented into a standalone program interfaced with various quantum chemistry packages, demonstrating its applicability on systems of increasing complexity.

Simulation of the distribution of astatine-211 solution dispersion in a lab room

Summary: The study aimed to evaluate the distribution of Astatine-211 (At-211) solution dispersion in a small animal cage using autoradiography imaging to simulate the dispersion of At-211 in a lab room to eventually prevent user's risk of internal exposure in terms of radiation safety. The mean activity of Free At-211 was 2.3 times higher than that of (NaAt)-At-211 on all autoradiography images. In the cage, the shape of the dispersion of (NaAt)-At-211 was almost homogeneous, whereas that of Free At-211 was more heterogeneous. The At-211 solution vaporized naturally and was distributed heterogeneously in the cage, and the chemical properties of At-211 influenced their behaviors, which must be considered to minimize the risks of radiation safety.
