Liquid-state NMR spectroscopy for complex carbohydrate structural analysis: A hitchhiker's guide

Immacolata Speciale et al.

Summary: This article provides a practical guide to the structural determination of carbohydrates using liquid-state NMR. The narrow range of chemical shifts in the NMR signals of carbohydrates makes it challenging to identify individual monosaccharide units. However, attributing the NMR chemical shifts can reveal the nature of the constituents, their interconnections, and even their absolute configuration, paving the way for more sophisticated analyses.

Structure, substrate recognition and initiation of hyaluronan synthase

Finn P. Maloney et al.

Summary: In this study, we investigated the substrate binding and polymer synthesis of hyaluronan synthase (HAS) using cryo-electron microscopy structures, biochemical analyses, and molecular dynamics simulations. Our findings provide detailed insights into the transmembrane process of HAS, including substrate selection, polymerization, and regulation of hyaluronan synthesis. This research is important for understanding the biosynthesis of acidic extracellular polysaccharides and one of the most abundant glycosaminoglycans in the human body.

N-glycans from Paramecium bursaria chlorella virus MA-1D: Re-evaluation of the oligosaccharide common core structure

Immacolata Speciale et al.

Summary: This study analyzed the N-glycans of Paramecium bursaria chlorella virus MA-1D and found that they share similarities with other chloroviruses, but lack a xylose unit believed to be part of a conserved core region. The study also discovered a gene with strong homology to a putative xylosyltransferase in reference chloroviruses, but with a premature stop codon mutation.

Chlorovirus PBCV-1 Multidomain Protein A111/114R Has Three Glycosyltransferase Functions Involved in the Synthesis of Atypical N-Glycans

Eric Noel et al.

Summary: The structures of N-linked glycans from the major capsid protein of chlorovirus PBCV-1 are unique and not found in other domains of life. A conserved central glycan core is shared among all known chloroviruses, with the protein A111/114R likely responsible for the attachment of three conserved residues in this core. This study suggests that chloroviruses use virus-encoded enzymes for glycosylation rather than the host cellular machinery.

The structure of a virus-encoded nucleosome

Marco Igor Valencia-Sanchez et al.

Summary: The cryo-EM structure of DNA-assembled histone pairs from Marseillevirus, a nucleocytoplasmic large DNA virus, reveals that they form viral nucleosomes with highly conserved features similar to canonical eukaryotic nucleosomes. Viral histones from Marseillevirus form 'forced' heterodimers that assemble into tetramers, which in turn assemble DNA to form structures virtually identical to canonical eukaryotic nucleosomes.

Identification of a Chlorovirus PBCV-1 Protein Involved in Degrading the Host Cell Wall during Virus Infection

Summary: Chloroviruses infect eukaryotic organisms by penetrating the host's cell wall, with protein A561L(D4) playing a key role in degrading the cell wall. This protein is present in all 52 genomically sequenced chloroviruses and can increase the specific infectivity of PBCV-1.

Expanding the Occurrence of Polysaccharides to the Viral World: The Case of Mimivirus

Anna Notaro et al.

Summary: The traditional perception of viruses being small and hijacking host machinery for glycosylation is challenged by the discovery of giant viruses with complex genomes and unique polysaccharide glycocalyces. These findings blur the boundaries between giant viruses and the cellular world, opening new avenues in the field of viral glycobiology.

Characterization of an aminotransferase from Acanthamoeba polyphaga Mimivirus

Chase A. Seltzner et al.

Summary: Acanthamoeba polyphaga Mimivirus is a complex virus that infects amoeba, with its DNA genome encoding nearly 1,000 proteins, including enzymes for the biosynthesis of unusual sugars. The enzyme L136 in Mimivirus is responsible for installing the amino group in the sugar, showing differences in catalytic efficiency compared to the structurally related DesI from Streptomyces venezuelae. The structure of L136 provides new information on virally encoded PLP-dependent enzymes and sugar aminotransferases.

A Persistent Giant Algal Virus, with a Unique Morphology, Encodes an Unprecedented Number of Genes Involved in Energy Metabolism

Romain Blanc-Mathieu et al.

Summary: Recent studies challenging the traditional view of viruses as entities with limited metabolic capabilities have identified metabolic genes in viruses with large genomes, such as Prymnesium kappa virus RF01 (PkV RF01). This virus encodes numerous proteins involved in energy metabolism and exhibits unique phenotypic features compared to other viruses in the Mimiviridae family. The presence of these metabolic genes suggests that viruses may actively regulate host energy metabolism and have complex evolutionary histories.

Synthesis of the hyper-branched core tetrasaccharide motif of chloroviruses

Bijoyananda Mishra et al.

Summary: The chemical synthesis of complex oligosaccharides, especially those with hyper-branched structures and specific glycosidic bonds, is a challenging task due to factors such as reactivity of glycosyl donors and acceptors, and proper tuning of solvent/temperature/activator. In this study, a semi-conserved hyper-branched core tetrasaccharide motif was successfully synthesized from chloroviruses, involving a thorough study of various factors to achieve a stereoselective galactopyranosidic linkage.
