Microtubule Stabilization Promotes Microcirculation Reconstruction After Spinal Cord Injury

Yang-Yang Duan et al.

Summary: The study demonstrates that microtubule stabilization treatment can improve microcirculation reconstruction after spinal cord injury by increasing the number of microvessels, pericytes, and perfused microvessels, while also reducing apoptosis and migration of endothelial cells and pericytes and promoting their proliferation and survival. This suggests the potential of microtubule stabilization as a therapeutic target to reduce microcirculation dysfunction after SCI in the clinic.

Sulfated glycans engage the Ang-Tie pathway to regulate vascular development

Matthew E. Griffin et al.

Summary: Heparan sulfate glycosaminoglycans (HS GAGs) regulate Ang-Tie signaling by interacting directly with Ang ligands and Tie1 receptors, enhancing endothelial survival signaling and maintaining Tie1 stability within the mature vasculature. Loss of HS-Tie1 binding leads to decreased Tie protein levels, pathway suppression, and aberrant retinal vascularization.

Single-Cell Analysis of Blood-Brain Barrier Response to Pericyte Loss

Maarja A. Mae et al.

Summary: Through single-cell RNA sequencing and other methods, the study revealed the transcriptional, morphological, and functional changes in brain endothelial cells in response to pericyte deficiency. Endothelial cells without pericyte contact maintain a BBB-specific gene expression profile and acquire a venous-shifted molecular pattern.

Conserved and context-dependent roles for pdgfrb signaling during zebrafish vascular mural cell development

Koji Ando et al.

Summary: The study demonstrates that Pdgfrb signaling plays a critical role in mural cells in zebrafish, and its absence can lead to issues such as cranial hemorrhage and vessel dilation. Similar to mice, pdgfrb mutant zebrafish also show defects in the structure of glomerulus, but normal development of hepatic stellate cells.

Local delivery of USC-derived exosomes harboring ANGPTL3 enhances spinal cord functional recovery after injury by promoting angiogenesis

Yong Cao et al.

Summary: In this study, it was demonstrated that a locally administered injection of USC-Exo embedded in hydrogel can deliver ANGPTL3 to the injured spinal cord region. Additionally, the administration of human USC-Exo could enhance spinal cord neurological functional recovery by promoting angiogenesis. Mechanistic studies revealed that ANGPTL3 enriched in USC-Exo is required for their ability to promote angiogenesis through the PI3K/AKT signaling pathway.

The quiescent endothelium: signalling pathways regulating organ-specific endothelial normalcy

Nicolas Ricard et al.

Summary: Endothelial cells play a crucial role in controlling oxygen, nutrient exchange, and cellular trafficking between blood and organs. The quiescent state of endothelium is actively regulated and constantly receives important signalling inputs. Understanding organ-specific pathways and their disruption in vascular pathologies can lead to the development of preventive strategies and new therapeutic targets.

Exosomal miR-155 from M1-polarized macrophages promotes EndoMT and impairs mitochondrial function via activating NF-κB signaling pathway in vascular endothelial cells after traumatic spinal cord injury

Summary: The study revealed that M1-BMDMs promote EndoMT of vascular endothelial cells, exacerbating BSCB disruption after SCI and causing dysfunction of mitochondria and accumulation of ROS. Exosomal miR-155/SOCS6/p65 axis plays a crucial role in regulating EndoMT process and mitochondrial function in vascular endothelial cells.

Neuroinflammatory astrocyte subtypes in the mouse brain

Philip Hasel et al.

Summary: This study investigated the transcriptome changes of mouse cortical astrocytes after an acute inflammatory stimulus, revealing rapid and drastic changes over time. By sequencing thousands of astrocytes at single-cell resolution, it was shown that inflammation causes a widespread response with distinct subtypes undergoing inflammatory transitions with defined transcriptomic profiles. Spatial transcriptomics and in situ hybridization were used to attribute specific inflammation-induced reactive astrocyte sub-states to certain brain regions.

Hypoxia response element-directed expression of bFGF in dental pulp stem cells improve the hypoxic environment by targeting pericytes in SCI rats

Sipin Zhu et al.

Summary: DPSCs expressing bFGF under the regulation of 5HRE were found to differentiate into CD13(+) pericytes and upregulate N-cadherin expression, promoting their re-attachment to vascular endothelial cells. This led to an increased flow rate of blood in microvessels and improved the local hypoxia microenvironment in SCI.
