Itraconazole attenuates the stemness of nasopharyngeal carcinoma cells via triggering ferroptosis

Ying Xu et al.

Summary: Radiotherapy is commonly used for treating nasopharyngeal carcinoma, but radioresistance is a significant issue in clinical practice. This study found that nasopharyngeal carcinoma spheroids exhibit resistance to both radiotherapy and ferroptosis. Itraconazole treatment triggered ferroptosis in these spheroids, reducing cell viability and partially reversing radioresistance by sequestering iron in lysosomes.

Lysosome Fe2+ release is responsible for etoposide- and cisplatin-induced stemness of small cell lung cancer cells

Kangwu Wang et al.

Summary: This study demonstrates that etoposide and cisplatin can induce stemness in small cell lung cancer cells through the HIF/Ferritin axis, playing a crucial role in tumor progression and recurrence. The mechanism of iron's involvement in chemotherapy-induced stemness, regulated by the HIF/Ferritin axis, is essential for understanding the impact on tumor development.

A Modular Synthetic Route Involving N-Aryl-2-nitrosoaniline Intermediates Leads to a New Series of 3-Substituted Halogenated Phenazine Antibacterial Agents

Hongfen Yang et al.

Summary: The study introduced a new series of halogenated phenazines (HP) with potent antibacterial and biofilm-killing activities, targeting and eradicating planktonic and biofilm bacteria, showing efficacy against methicillin-resistant Staphylococcus aureus and Mycobacterium tuberculosis. Transcriptional analysis revealed rapid iron starvation induced by HPs in MRSA biofilms, offering new insights into treating challenging infections._HP 29 demonstrated efficacy in dorsal wound infection models in mice, suggesting HPs could lead to significant advances in infection treatment.

Chemistry and biology of ferritin

Marina Plays et al.

Summary: Iron is an essential element for cells, with ferritin playing a key role in iron storage and cell metabolism, as well as its link to ferroptosis. The chemistry of iron, including its oxidation states, is crucial for its biological functions and reactivity, particularly in the context of ferritin biology. Ferritin localization within cells and its function in tissue-specific contexts are tightly interlinked, and disruptions in iron homeostasis are implicated in diseases like cancer and COVID-19. Ferritin serves as a potential biomarker, with ongoing research exploring its applications in imaging and drug delivery.
