Aggravation of pulmonary fibrosis after knocking down the aryl hydrocarbon receptor in the insulin-like growth factor 1 receptor pathway

Summary: This study investigated the cascade pathway involving IGF1R and AhR in the development of idiopathic pulmonary fibrosis. The results showed increased IGF1R levels and decreased AhR expression in idiopathic pulmonary fibrosis patients and animal models. Knockout of AhR aggravated lung fibrosis, while the use of IGF1R inhibitor and AhR agonist attenuated such effects. TGF-beta1 and BLM suppressed AhR expression through endoplasmic reticulum stress and activated IGF1R. This study provides a potential target for the treatment of lung injury and fibrosis.

Summary: This study reveals that DNA methylation and overexpression of MBD2 are associated with fibroblast differentiation and pulmonary fibrosis in idiopathic pulmonary fibrosis (IPF) and a mouse model. TGF-β1 induces global DNA hypermethylation and Mbd2 overexpression in a TβRI/Smad3 dependent manner, leading to repression of Erdr1 expression and promotion of fibroblast differentiation into myofibroblasts. Enhancing Erdr1 expression can significantly reverse pulmonary fibrosis.

Summary: Treatments for pulmonary fibrosis are currently ineffective due to unclear molecular pathogenesis and therapeutic targets. However, a study has shown that disturbed LDL-LDLR metabolism may contribute to the dysfunction of endothelial and epithelial cells, and fibroblasts during pulmonary fibrogenesis. Pharmacological restoration of LDLR levels could potentially have beneficial effects on pulmonary fibrosis.

Summary: A simplified repetitive bleomycin instillation strategy was developed to mimic persistent and progressive pulmonary fibrosis, recapitulating histological and radiographic characteristics of idiopathic pulmonary fibrosis (IPF). Compared to the single bleomycin instillation model, this simplified repetitive model may be better suited for addressing mechanistic questions about IPF pathogenesis and preclinical studies of antifibrotic drug candidates.
