ApoE Cascade Hypothesis in the pathogenesis of Alzheimer's disease and related dementias

Yuka A. Martens et al.

Summary: The epsilon 4 allele of the apolipoprotein E gene is a strong genetic risk factor for Alzheimer's disease and other neurodegenerative conditions. The small amino acid differences in the sequence among the three isoforms of apoE have profound effects on aging and disease-related pathways. This study proposes a novel ApoE Cascade Hypothesis that suggests the biochemical and biophysical properties of apoE impact a cascade of events, ultimately leading to aging-related pathogenic conditions including Alzheimer's disease.

Cholesterol and matrisome pathways dysregulated in astrocytes and microglia

Julia Tcw et al.

Summary: The impact of APOE4 on human brain cellular function remains unclear. This study investigates the mechanisms by which APOE4 affects brain cells and reveals dysregulation of cholesterol metabolism and extracellular matrix in astrocytes, which may contribute to increased risk of AD.

Tackling neurodegenerative diseases with genomic engineering: A new stem cell initiative from the NIH

Daniel M. Ramos et al.

Summary: iNDI is the largest iPSC genome engineering project, aiming to model over 100 mutations associated with Alzheimer's disease and related dementias. The resulting cell lines and phenotypic datasets will be widely shared with the research community.
