Antiviral Strategies Using Natural Source-Derived Sulfated Polysaccharides in the Light of the COVID-19 Pandemic and Major Human Pathogenic Viruses

Bimalendu Ray et al.

Summary: Currently available antiviral drugs can only target a small fraction of human pathogenic viruses. The outbreak of SARS-CoV-2 has emphasized the urgent need for molecules that can quickly treat novel or re-emerging viral infections. Sulfated polysaccharides, which are found on the surfaces of both host cells and human viruses, have shown high and sometimes broad-spectrum antiviral activity. Several new polysaccharide-derived drugs are currently being investigated.

Analysis of celluloses, plant holocelluloses, and wood pulps by size-exclusion chromatography/multi-angle laser-light scattering

Yuko Ono et al.

Summary: Size-exclusion chromatography with multi-angle laser-light scattering and refractive index detection (SEC/MALLS/RI) provides detailed information on the number- and weight-average molar masses, molar mass distributions, conformations, and linear/branched structures of polymers. By dissolving in a solvent system similar to 8% (w/w) LiC1/DMAc, analysis can be conducted on pure celluloses, chemical wood pulps, and plant holocelluloses. The results vary depending on the cellulose and holocellulose resources, with linear and random-coil conformations observed in pure celluloses and high-molar-mass cellulose fractions. Softwood holocellulose and bleached kraft pulp, on the other hand, exhibit alkali-stable cellulose/glucomannan branched structures in the high-molar-mass fractions.

Controlled sulfation of mixed-linkage glucan by Response Surface Methodology for the development of biologically applicable polysaccharides

Laleh Solhi et al.

Summary: Endogenous and exogenous sulfated polysaccharides have potent biological activities, and controlled chemical sulfation of alternative polysaccharides, such as mixed-linkage beta(1,3)/beta(1,4)-glucan (MLG), holds promise to overcome limited availability and heterogeneity of naturally sulfated polysaccharides. By optimizing the degree-of-substitution (DS) externally validated through production of sulfated MLGs (S-MLGs), this study demonstrates a route to medically important polymers from an economical agricultural polysaccharide.
