Nanobodies as molecular imaging probes

Sarah Barakat et al.

Summary: Camelidae derived single-domain antibodies, known as nanobodies, are small antibody fragments with full antigen-binding capacity. They have gained popularity due to their small size, high specificity, strong affinity, excellent stability, and modularity. Nanobodies are now widely used as molecular probes in the field of molecular imaging, allowing the detection and tracking of various targets. This review provides an overview of their applications and discusses current techniques for their generation and delivery.

An AKT2-specific nanobody that targets the hydrophobic motif induces cell cycle arrest, autophagy and loss of focal adhesions in MDA-MB-231 cells

Tijs Merckaert et al.

Summary: The AKT kinase family is a prominent target for cancer therapy, with the three isoforms of AKT having distinct and sometimes opposing functions. By utilizing AKT2-specific nanobodies, researchers were able to modulate AKT2 and inhibit proliferation in breast cancer cells, affecting phosphorylation of downstream proteins and resulting in cell cycle arrest and induction of autophagy. These findings suggest that targeting AKT2 and its hydrophobic motif could be a viable approach for cancer therapy.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Self-Assembled Nanobodies as Selectively Targeted, Nanostructured, and Multivalent Materials

Laura Sanchez-Garcia et al.

Summary: Nanobodies are valuable tools in advanced therapeutic strategies, but their small size and limited valence for interactions may pose restrictions for in vivo applications. Modular protein constructs have been designed to fuse nanobodies with protein domains, promoting self-assembly into stable nanoparticles with maintained specificity for targets.
