Clinical relevance of blood-based ctDNA analysis: mutation detection and beyond

Laura Keller et al.

Summary: Cell-free DNA derived from tumors can be detected in the plasma of cancer patients, and significant progress has been made in the detection of mutations in circulating tumor DNA. In addition to mutation detection, cfDNA can be used to analyze other features of DNA, providing new avenues for liquid biopsy analysis.

The role of HPV-induced epigenetic changes in cervical carcinogenesis (Review)

Martha Laysla Ramos Da Silva et al.

Summary: Cervical cancer, caused by high-risk HPV infection, remains the second most common cancer type in women in developing regions. Epigenetic modifications are stable changes in DNA that affect gene expression and genomic stability, playing a key role in various biological processes. These modifications have been identified in both viral DNA and infected cell genomes, contributing to the development of cervical cancer.

Clinical implications of plasma circulating tumor DNA in gynecologic cancer patients

Lindsey M. Charo et al.

Summary: The molecular characterization of gynecologic cancers through ctDNA analysis plays a vital role in determining prognosis and guiding personalized therapy. This study found that higher ctDNA mutation frequency is associated with poor overall survival, while therapy tailored to ctDNA alterations leads to improved outcomes. The concordance between tissue and ctDNA genomic results remains high, demonstrating the potential utility of ctDNA in individualizing cancer therapy for gynecologic cancer patients.

Sanjana Balachandra et al.

Summary: This study found that HPV-16 E6 antibodies and circulating HPV-16 DNA are the most representative and promising blood-based biomarkers for HPV-associated cancers. Comparative validity analyses are needed, and further research on biomarkers targeting more high-risk HPV types is warranted.

Association between Gene Promoter Methylation and Cervical Cancer Development: Global Distribution and A Meta-analysis

Aissam El Aliani et al.

Summary: DNA methylation is a key epigenetic event related to gene silencing and carcinogenesis. The meta-analysis evaluated the association between promoter methylation of specific genes and the risk of cervical cancer development, finding higher methylation frequencies in certain genes in cervical cancer cases compared to controls. Promoter methylation could potentially serve as a promising noninvasive biomarker for early cervical lesions.

Summary: The study conducted a ctDNA-based analysis on over 10,000 pan-cancer Chinese patients, revealing age-related CH variants, detection rates of ctDNA in different cancer types, and the frequencies of potential driver genes. The findings may be valuable for identifying therapeutic targets and developing combined treatment strategies.

Summary: A study in England found a significant reduction in cervical cancer and CIN3 incidence in young women after the introduction of the HPV immunisation programme, especially in those vaccinated at age 12-13. The programme has successfully almost eliminated cervical cancer in women born since September 1, 1995.

Liquid Biopsy in Cervical Cancer: Hopes and Pitfalls

Paola Cafforio et al.

Summary: Cervical cancer is the fourth most common cancer in women globally, with 90% of cancer-related deaths occurring in developing countries. Liquid biopsy may serve as an alternative (or complementary) tool for screening, diagnosis, and follow-up of cervical cancer, including the detection and isolation of circulating tumor cells, cell tumor DNA sequencing, coding and non-coding RNA detection, and exosomal miRNA identification. Further validation and standardization of these methodologies are needed before clinical use.

HPV ctDNA detection of high-risk HPV types during chemoradiotherapy for locally advanced cervical cancer

L. Cabel et al.

Summary: The study designed HPV droplet digital PCR (ddPCR) assays for blood detection and monitored blood HPV circulating tumor DNA (HPV ctDNA) levels in patients with LACC undergoing CRT. Results showed successful detection of HPV ctDNA in 69% of patients before CRT, with a correlation between HPV-ctDNA levels and HPV copy number in tumors. Detection of HPV-ctDNA before CRT was associated with tumor stage and lymph node status, while positive detection during follow-up was linked to lower disease-free and overall survival rates.

Summary: The study showed that mutant genes identified in plasma DNA samples from MRCC patients influence treatment outcomes, with a decrease in mutations after chemotherapy associated with better response. Longitudinal monitoring with ctDNA in liquid biopsy samples provides predictive and prognostic information during treatment.
