The complete sequence of a human genome

Sergey Nurk et al.

Summary: The Telomere-to-Telomere (T2T) Consortium has presented a complete sequence of a human genome, T2T-CHM13, which covers the whole genome except for the Y chromosome. This new sequence includes gapless assemblies, error corrections in previous references, and nearly 200 million base pairs of additional sequence with gene predictions, including protein coding genes. The completion of important regions allows for further studies on genetic variations and functions.

Genetic variation at mouse and human ribosomal DNA influences associated epigenetic states

Francisco Rodriguez-Algarra et al.

Summary: This study reveals the existence of genetic haplotypes in rDNA units of C57BL/6J mice, which influence the epigenetic state and transcriptional output of these units. However, human rDNA units show limited evidence of genetic haplotypes. Additionally, adjacent units in different species show similar epigenetic profiles, and the overall epigenetic state at rDNA is strongly positively correlated with the total rDNA copy number.

The human ribosomal DNA array is composed of highly homogenized tandem clusters

Yutaro Hori et al.

Summary: Recent studies have revealed that human cells have a relatively regular rDNA structure, with variations in the noncoding regions of each rDNA copy. Nanopore sequencing further showed that all noncoding regions are heavily methylated, while about half of the coding regions are clearly unmethylated.

Gene body methylation safeguards ribosomal DNA transcription by preventing PHF6-mediated enrichment of repressive histone mark H4K20me3

Xiaoke Huang et al.

Summary: Gene body methylation is positively correlated with rRNA gene transcription, mainly maintained by DNMT1, deficiency or downregulation of which leads to decreased methylation levels and increased enrichment of PHF6 on gene bodies, ultimately inhibiting rDNA transcription.
