Barley Anther and Meiocyte Transcriptome Dynamics in Meiotic Prophase I

Summary: In this study, the temporal dynamics of transcript abundance in barley anthers and meiocytes were investigated using RNA-seq, revealing significant transcriptional changes in anthers at the transition from pre-meiosis to leptotene-zygotene and remarkable stability in meiocytes throughout prophase I after the initial reprogramming at meiosis entry. Additionally, only 24% of putative meiotic gene orthologs showed differential transcript abundance, indicating the complexity of regulatory networks involved in early meiotic processes.

The results showed that lncRNAs play a critical role in pollen mother cells meiosis, anther and pollen development during the fertility transition. Some lncRNAs were identified as precursors for miRNAs or endogenous target mimics, regulating the expression of miRNAs and mRNAs and contributing to the fertility transition process.

Gene Ontology and pathway analysis showed that the target genes of these miRNAs were enriched in specific pathways, particularly in Hepatitis B and cancer pathways. Further analysis using a protein-protein interaction network identified 10 miRNAs predicted to play crucial roles in the growth and development of goats, particularly in disease occurrence and prevention.
