Biophysical and functional properties of purified glucose-6-phosphatase catalytic subunit 1

Derek P. Claxton et al.

Summary: Glucose-6-phosphatase catalytic subunit 1 (G6PC1) plays a crucial role in regulating blood glucose levels and its dysregulation is linked to diabetes and glycogen storage disease type 1a. However, limited understanding of G6PC1 structure and mechanism is due to challenges in expression and purification. In this study, researchers successfully analyzed the catalytically active G6PC1 using biophysical and biochemical tools, providing a foundation for future studies on the structural biology and role of G6PC1 in complex metabolic disorders.

Time-Dependent Efficacy of Checkpoint Inhibitor Nivolumab: Results from a Pilot Study in Patients with Metastatic Non-Small-Cell Lung Cancer

Abdoulaye Karaboue et al.

Summary: The study found that administering nivolumab in the morning was nearly four times more effective for metastatic non-small-cell lung cancer (NSCLC) patients compared to afternoon dosing, regardless of patient characteristics. Randomized trials are needed to further identify the optimal timing for checkpoint inhibitors.

Post-transcriptional circadian regulation in macrophages organizes temporally distinct immunometabolic states

Emily J. Collins et al.

Summary: The circadian clock plays a crucial role in immunity, with post-transcriptional regulation influencing macrophage clock regulatory output significantly, particularly in metabolic pathways. Daily variations in metabolic pathways, ATP production, mitochondrial morphology, and phagocytosis correspond to post-transcriptional temporal-gating and are essential for determining the response of macrophages to immune stimuli.

Molecular mechanism of the repressive phase of the mammalian circadian clock

Xuemei Cao et al.

Summary: The mammalian circadian clock is regulated by a transcription-translation feedback loop involving CLOCK-BMAL1 activators and CRY-PER repressors. Previous studies have shown that CRY and CRY-PER inhibit CLOCK-BMAL1 in different mechanisms. This study reveals the involvement of CRY-PER in displacing CLOCK-BMAL1 by recruiting CK1 delta, leading to the dissociation of CLOCK-BMAL1 from the E-box.

Identifying and Characterizing Serious Adverse Drug Reactions Associated With Drug-Drug Interactions in a Spontaneous Reporting Database

Lara Magro et al.

Summary: The study found that about one-third of patients exposed to potential drug-drug interactions actually experienced serious adverse drug reactions, and that a spontaneous reporting database can be used to identify and describe ADRs caused by drug-drug interactions. Among these ADRs, warfarin was the most frequently reported interacting drug and the most common ADRs were gastrointestinal or cerebral hemorrhagic events.

Clinical Implications of Combinatorial Pharmacogenomic Tests Based on Cytochrome P450 Variant Selection

Michael Sayer et al.

Summary: Although pharmacogenomics (PGx) has the potential to improve patient outcomes, its application is not yet routine. A study evaluated the detection rate of currently available combinatorial PGx (CPGx) tests based on cytochrome P450 (CYP) gene variants, showing significant variation across ethnic groups. Therefore, the patient's ethnic background should be carefully considered in selecting CPGx tests.
