Summary: This study developed an in vitro optimized process to prepare uniform and nucleic acid free HBc-based VLPs, successfully obtaining two types of VLPs through a disassembly-purification-reassembly procedure. It was found that the degree of disassembly had a significant impact on the protein yield, nucleic acid removal, and reassembly efficiency of the VLPs.
