Summary: Mitochondria, ancient bacterial endosymbionts of eukaryotic cells, have their own circular DNA and protein translation system. Recent advancements in cryoelectron microscopy and mass spectrometry have revealed the evolutionary divergence of mitochondrial ribosomes from their bacterial ancestors and their adaptation for synthesizing mitochondrial DNA encoded oxidative phosphorylation complex subunits. Furthermore, assembly pathway maps of mitoribosomes and associated factors have started to emerge, shedding light on the complex assembly process.

A conserved rRNA switch is central to decoding site maturation on the small ribosomal subunit

Summary: The study used single-particle cryo-electron microscopy to observe the effects of bacterial ribosome assembly factors on the conformation of the 30S ribosomal subunit, revealing a conserved secondary structure switch in the 16S ribosomal RNA central to decoding site maturation, and indicating the sequential order of action and molecular mechanisms for the assembly factors in coordinating and controlling this switch. Structural and mechanistic parallels between bacterial and eukaryotic systems suggest common folding features inherent to all ribosomes.
