Reprogramming of glucose metabolism of cumulus cells and oocytes and its therapeutic significance

Shogo Imanaka et al.

Summary: This review summarizes the molecular mechanisms of glucose metabolism during oocyte maturation, focusing on the role of pyruvate dehydrogenase (PDH) and the metabolic balance between aerobic glycolysis and the TCA cycle/OXPHOS. The understanding of the PDK-PDH axis is crucial for developing novel therapeutic strategies to improve oocyte quality.

Glucose metabolism characterization during mouse in vitro maturation identifies alterations in cumulus cells

Nazli Akin et al.

Summary: The novel two-step IVM method CAPA-IVM has improved embryology outcomes by synchronizing oocyte nuclear and cytoplasmic maturation, although there is still a noticeable efficiency gap compared with traditional IVF. Glucose metabolism studies have revealed metabolic shortcomings within CAPA-IVM COCs, with specific alterations in mouse COC glucose metabolism characterized for the first time. Future optimization strategies for CAPA-IVM could focus on adjusting culture media energy substrate concentrations and/or implementing co-culture strategies.

HYPOXIA AND REPRODUCTIVE HEALTH Hypoxia and ovarian function: follicle development , ovulation, oocyte maturation

Megan Lim et al.

Summary: The low oxygen environment within the ovarian follicle promotes oocyte development and survival through the activation of hypoxia-inducible factors (HIFs). Angiogenesis, regulated by HIF, plays a crucial role in follicle development and corpus formation. Ovulation is linked to HIF activity, with the luteinising hormone surge increasing HIF expression.

Reversing complete mechanical transzonal projections disruption during mouse in vitro follicle culture with unaltered oocyte competence

Anamaria-Cristina Herta et al.

Summary: The study shows that oocytes from cultured mouse follicles can overcome complete denudation, restoring functional transzonal projections, with no significant differences in meiotic and developmental competence compared to those from intact cultured follicles. These findings provide good premises for developing successful strategies to rescue human oocyte competence in in vitro culture approaches.
