Immunogenicity and humanization of single-domain antibodies

Martin A. Rossotti et al.

Summary: Single-domain antibodies have desirable properties as biologic drugs, but their sequences may increase immunogenicity risks. While most clinical trials show minimal immunogenicity, special caution is needed for highly potent sdAbs. Further research is needed to establish humanization thresholds to maximize human sequence content without compromising binding affinity or stability.

Antibody variable sequences have a pronounced effect on cellular transport and plasma half-life

Algirdas Grevys et al.

Summary: The variable region sequences of IgG antibodies significantly impact their cellular uptake, recycling, and degradation, as well as their binding to FcRn. Differences in charge patch also modulate binding and transport properties. Fc-engineering may improve FcRn binding and overcome unfavorable charge patches for longer plasma half-life.

Assessment of immunogenicity and drug activity in patient sera by flow-induced dispersion analysis

Morten E. Pedersen et al.

Summary: Biopharmaceuticals have transformed the treatment of various diseases, but they can also trigger immune responses that affect treatment efficacy and patient safety. Current immunogenicity assays have limitations in reliable and continuous monitoring. In this study, a new method called Flow Induced Dispersion Analysis (FIDA) was introduced for assessing immunogenicity and drug activity. The method provides a quantitative measurement under biologically relevant conditions.

In vitro discovery of a human monoclonal antibody that neutralizes lethality of cobra snake venom

Line Ledsgaard et al.

Summary: This study describes the discovery and optimization of a recombinant human monoclonal immunoglobulin G antibody against alpha-cobratoxin using phage display technology. The optimized antibody showed increased neutralization potency and efficacy, providing a promising prospect for the development of recombinant antivenoms based on human monoclonal or oligoclonal antibodies.

A super-potent tetramerized ACE2 protein displays enhanced neutralization of SARS-CoV-2 virus infection

Ami Miller et al.

Summary: A novel protein engineering technology has been used to produce a tetravalent ACE2-Fc-TD protein that effectively blocks SARS-CoV-2 virus infection. This tetrameric protein complex acts as a powerful decoy protein, capable of binding to the virus and preventing its invasion.

Multivalency transforms SARS-CoV-2 antibodies into ultrapotent neutralizers

Edurne Rujas et al.

Summary: By using human apoferritin protomer as a modular subunit, researchers have developed exceptionally potent neutralizers targeting SARS-CoV-2. Combining three different antibody specificities and the fragment crystallizable (Fc) domain on a single multivalent molecule enabled the platform to overcome viral sequence variability and deliver ultrapotent neutralizers against SARS-CoV-2. This MULTi-specific, multi-Affinity antiBODY (Multabody or MB) platform leverages binding avidity and multi-specificity to provide broad neutralization against SARS-CoV-2.

Universal endogenous antibody recruiting nanobodies capable of triggering immune effectors for targeted cancer immunotherapy

Haofei Hong et al.

Summary: UEAR Nbs, engineered universal endogenous antibody-recruiting nanobodies, are able to recruit endogenous IgGs onto the cancer cell surface in vitro, triggering potent immune responses to kill cancer cells. They have significantly improved half-lives in vivo and demonstrated antitumor efficacy in a murine model of EGFR positive triple-negative breast cancer (TNBC).
