Spatial organization of Dps and DNA-Dps complexes

Evgeniy Dubrovin et al.

Summary: The co-crystallization of DNA with Dps family proteins is essential for preserving bacterial DNA, but the spatial organization of DNA in DNA-Dps co-crystals and the interaction mechanism between DNA and Dps molecules require further investigation. The use of atomic force microscopy and small-angle X-scattering has provided insights into the structure of DNA-Dps complexes, shedding light on the non-specific nature of Dps binding with DNA and the arrangement of DNA along protein molecules on a Dps sheet.

Anomalous SAXS at P12 beamline EMBL Hamburg: instrumentation and applications

Andrey Yu Gruzinov et al.

Summary: Small-angle X-ray scattering (SAXS) is a method for studying nanostructured systems and biological macromolecules, while anomalous X-ray scattering (ASAXS) utilizes changes in scattering properties of atoms to obtain element-specific information. ASAXS measurements on biological macromolecules are challenging due to weak anomalous effects, but recent developments at the P12 beamline have allowed for successful ASAXS experiments on model systems.

Structural Rearrangement of Dps-DNA Complex Caused by Divalent Mg and Fe Cations

Liubov Dadinova et al.

Summary: Two independent methods, SAXS and cryo-EM, were used to investigate the effect of Mg2+ and Fe2+ cations on the Dps-DNA complex structure. It was found that Mg2+ ions block the interaction between Dps and DNA, while the presence of Fe2+ leads to complex destruction and aggregation without restoration. Cryo-EM analysis also revealed the presence of iron nanoparticles inside the Dps cavity next to acidic pore.
