Advances allowing feasible pyrG gene editing by a CRISPR-Cas9 system for the edible mushroom Pleurotus eryngii

Tingli Wang et al.

Summary: A study identified a highly efficient gene editing system in the edible mushroom Pleurotus eryngii, which can generate various insertions and deletions through non-homologous end joining and homology-directed repair. By using a point-mutated variant of the endogenous sdhB gene as a selection marker, the system was successfully established for the first time, potentially opening up new possibilities for improving edible mushroom cultivars.

Functional analyses of Pleurotus ostreatus pcc1 and clp1 using CRISPR/Cas9

Tatpong Boontawon et al.

Summary: This study investigated the molecular mechanisms controlling dikaryon formation in Agaricomycetes, specifically the role of A and B mating-type loci. Results showed that mutations in pcc1 in Coprinopsis cinerea can activate the A-regulated pathway, leading to pseudoclamp formation. Phenotypic analyses of Popcc1 and Poclp1 mutants in Pleurotus ostreatus suggest conserved and diverse mechanisms in sexual development among Agaricomycetes.
