Critical Pressure of Intramural Delamination in Aortic Dissection

Ehsan Ban et al.

Summary: Computational models are used to study the mechanisms of aortic dissection, and the relationship between pressure and geometric/mechanical factors are analyzed to reveal the patterns of initiation and propagation of aortic dissection. Further experimental data is needed to validate and expand these results.

mTOR inhibition prevents angiotensin II-induced aortic rupture and pseudoaneurysm but promotes dissection in mice

Changshun He et al.

Summary: This study investigates the role of mTOR signaling in aortic disease. The activation of mTOR in vascular wall cells determines whether aortic tears progress to dissection or rupture. Inhibition of mTOR signaling prevents aortic rupture but promotes dissection. Thus, caution should be exercised when exploring mTOR inhibition for aortic disease.

Differential propensity of dissection along the aorta

Ehsan Ban et al.

Summary: Research shows that aortic dissections progress through delamination of the aortic wall, with two distinct modes of tearing observed in different regions of the aorta. A phase-field finite-element model was implemented to study the correlation between tearing pressure and local geometry and material properties. The presence of thick interlamellar radial struts in the abdominal region was found to increase tearing pressure and provide insights into the mechanical factors influencing aortic dissections.

Transforming Growth Factor-β and the Renin-Angiotensin System in Syndromic Thoracic Aortic Aneurysms: Implications for Treatment

Daan C. H. van Dorst et al.

Summary: Thoracic aortic aneurysms (TAAs) are permanent pathological dilatations of the thoracic aorta that can lead to life-threatening complications, frequently occurring in individuals with genetic predispositions such as Marfan syndrome and Loeys-Dietz syndrome. Research indicates the significant role of TGF-beta signaling and the renin-angiotensin system in the pathology of TAAs, with ongoing studies exploring the therapeutic potential of ARBs and beta-blockers in syndromic TAA patients.

Insights on the Pathogenesis of Aneurysm through the Study of Hereditary Aortopathies

Tyler J. Creamer et al.

Summary: Thoracic aortic aneurysms (TAA) are permanent dilations of the aorta that predispose patients to life-threatening aortic dissection or rupture. Pathogenic variants causing hereditary TAA compromise molecular processes essential for aortic homeostasis. These variants impair signal transduction and signaling pathways involved in the modulation of vascular smooth muscle cells (VSMCs) responses to mechanical and biochemical cues from their environment, ultimately leading to vessel degeneration and mechanical failure.

Progressive Microstructural Deterioration Dictates Evolving Biomechanical Dysfunction in the Marfan Aorta

Cristina Cavinato et al.

Summary: This study investigates the relationship between mutations in the fibrillin-1 gene leading to Marfan syndrome and thoracic aortic aneurysms, using mouse models to show that as the condition worsens, elastic fiber defects, collagen fiber remodeling, and cell reorganization increase progressively.

Biomechanical consequences of compromised elastic fiber integrity and matrix cross-linking on abdominal aortic aneurysmal enlargement

D. Weiss et al.

Summary: This study investigated the progression of abdominal aortic aneurysms using a mouse model, revealing that loss of elastic fiber integrity is a strong initiator of aneurysms. The research also showed that glycosaminoglycan accumulation is pathologic, not adaptive, and heightened collagen deposition is ineffective if not cross-linked.
