Defining roles of specific reactive oxygen species (ROS) in cell biology and physiology

Helmut Sies et al.

Summary: This article discusses the key issues associated with identifying the physiological roles of oxidants, with a focus on H2O2 and O-2(.-). The article emphasizes the need for more precise measurement of oxidants and specific identification of their signaling targets. It also highlights the importance of inter-organellar communication and the contribution of environmental exposures. Development of tools for real-time detection and quantification of individual oxidants is crucial for achieving these goals.

Spectroscopic and theoretical evidences for the surface binding of voglibose drug with DNA

R. Durga Priyadharshini et al.

Summary: In this study, the binding of voglibose (VOG) with CT-DNA was investigated using various spectroscopic techniques. The results showed that the binding process is spontaneous and driven by hydrophobic and H-bonding interactions. Competitive displacement assays suggested that VOG binds on the surface of CT-DNA, which was further supported by viscosity measurements. Molecular simulation studies also confirmed the surface binding of VOG on DNA through hydrophobic interactions and H-bonding.

Synthesis, DNA-/bovine serum albumin-binding affinity, and cytotoxicity of dinuclear platinum(II) complexes with 1,6-naphthyridine-bridging ligand

Bata Konovalov et al.

Summary: The dinuclear platinum(II) complexes Pt1 and Pt2 were synthesized and characterized, with Pt1 showing higher cytotoxicity towards solid cancer cell lines and lower cytotoxicity towards mMSC cells compared to Pt2. Furthermore, Pt1 complex induced apoptosis in 4T1 and A549 cells by increasing pro-apoptotic caspase-3 expression and decreasing anti-apoptotic Bcl-2 and Ki-67 expression, suggesting its potential antitumor capacity through facilitating apoptosis and inhibiting tumor cell proliferation.

What blocks more anticancer platinum complexes from experiment to clinic: Major problems and potential strategies from drug design perspectives

Kun Peng et al.

Summary: Research on platinum coordination complexes as anticancer drugs has been conducted for over half a century with the inspiration from the success of cisplatin in cancer chemotherapy. Despite the emergence of numerous promising platinum complexes, most have failed to enter clinical applications. Strategies have been proposed in recent decades to address the complicated reasons for the abandonment of platinum antitumor candidates from experiment to clinic. With advancements in targeted therapy and immunotherapy, anticancer platinum drugs are facing new opportunities and challenges.
