Public health actions to control new SARS-CoV-2 variants Comment

Nathan D. Grubaugh et al.

Summary: Recent reports indicate that certain SARS-CoV-2 genetic variants, such as B.1.1.7, may be more transmissible and spreading rapidly worldwide. To prevent further exacerbation of the pandemic, it is necessary to increase surveillance and implement measures to reduce community transmission.

SARS-CoV-2 inhibits induction of the MHC class I pathway by targeting the STAT1-IRF1-NLRC5 axis

Ji-Seung Yoo et al.

Summary: The authors show that SARS-CoV-2 targets key transcriptional regulators of the MHC class I pathway to hinder antigen presentation, revealing an immune evasion mechanism that suppresses host immune response.

Piglet immunization with a spike subunit vaccine enhances disease by porcine epidemic diarrhea virus

Jieshi Yu et al.

Summary: Immunization with a vaccine containing PEDV spike protein induced high levels of neutralizing antibodies in mice and piglets, but resulted in worsened symptoms and increased virus replication in piglets. These findings highlight a previously unrecognized challenge in PEDV vaccine research with implications for coronavirus vaccine development.

Innate immune sensing of coronavirus and viral evasion strategies

Yusuke Kasuga et al.

Summary: Understanding how the innate immune system detects coronaviruses and how the viruses evade detection is crucial for developing novel strategies to combat infections. Constant evolution of coronaviruses to evade host immune responses underscores the need for in-depth research into specific viral proteins and mechanisms that suppress immunity. Targeting key receptors like TLR7 in the host immune system could be a promising therapeutic approach against coronavirus infections.

Betulonic Acid Derivatives Interfering with Human Coronavirus 229E Replication via the nsp15 Endoribonuclease

Annelies Stevaert et al.

Summary: This study identified a class of HCoV inhibitors targeting nsp15, with lead molecule 5h showing strong inhibitory effects on HCoV-229E replication. However, a mutant virus with nsp15 endoribonuclease active site mutation showed reduced sensitivity to 5h. The findings revealed a druggable pocket in the nsp15 hexamer with relevance for anti-coronavirus drug development.
