Metabolic adjustments of blood-stage Plasmodium falciparum in response to sublethal pyrazoleamide exposure

Summary: This study investigated the effects of pyrazoleamide drugs on the metabolism of Plasmodium falciparum. The researchers found that the drug treatment resulted in the adjustment of carbohydrate metabolism in the parasites, leading to enhanced myoinositol synthesis. This metabolic adaptation caused a decrease in the flux through the pentose phosphate pathway, resulting in a decreased rate of RNA synthesis and an increase in oxidative stress. The study suggests that enhanced myoinositol synthesis may be an important mechanism underlying the adaptation and emergence of resistance to pyrazoleamide drugs in P. falciparum exposed to sublethal doses.
